祛瘀生新方对轻中度溃疡性结肠炎小鼠胆汁酸代谢的影响

目的 观察祛瘀生新方对轻中度溃疡性结肠炎小鼠粪便中胆汁酸谱的影响,探讨其治疗溃疡性结肠炎的作用机制.方法 取22 只C57BL/6J小鼠,随机选择5 只作为空白组,其余小鼠采用自由饮用4%葡聚糖硫酸钠溶液方法建立轻中度溃疡性结肠炎模型.将造模成功小鼠随机分为模型组、祛瘀生新方组与美沙拉秦组,每组5 只,祛瘀生新方组与美沙拉秦组分别予相应药物灌胃,空白组和模型组给予蒸馏水灌胃,均1 次/d,连续14 d.观察各组小鼠活动、饮食、排便等情况;末次灌胃结束后采用靶向代谢组学技术分析小鼠粪便胆汁酸代谢情况,HE染色观察结肠组织病理形态.结果 模型组小鼠精神萎靡,体重减轻,毛发减少,便血,解稀便;结肠组织中有大量炎性细胞及中性粒细胞浸润.祛瘀生新方组及美沙拉嗪组小鼠灌胃结束时活动灵敏,毛发、饮食及排便无明显异常;结肠组织中有少许炎性细胞及中性粒细胞浸润.与空白组比较,模型组中熊脱氧胆酸(UDCA)、石胆酸(LCA)、牛磺-α-鼠胆酸(T-α-MCA)、牛磺猪胆酸(THCA)、甘氨猪胆酸(GH-CA)、熊果胆酸(UCA)、猪胆酸(HCA)、脱氧胆酸(DCA)、α-鼠胆酸(α-MCA)、牛磺鹅脱氧胆酸(TCDCA)、牛磺脱氧胆酸(TDCA)、甘氨猪脱氧胆酸(GHDCA)、猪去氧胆酸(HDCA)、甘氨石胆酸(GLCA)、牛磺胆酸(TCA)、胆酸(CA)含量均明显降低(P均<0.05),鹅脱氧胆酸(CDCA)、β-鼠胆酸(β-MCA)、甘氨胆酸(GCA)、脱氢胆酸(dehydro-CA)含量均明显升高(P均<0.05);与模型组比较,祛瘀生新方组和美沙拉嗪组UDCA、LCA、T-α-MCA、THCA、GHCA、UCA、α-MCA、HCA、DCA、TCDCA、TDCA、GHDCA、HDCA、GLCA、TCA、CA含量均明显升高(P均<0.05),CDCA、β-MCA、GCA、dehydeo-CA含量均明显降低(P均<0.05).结论 轻中度溃疡性结肠炎小鼠粪便中胆汁酸谱发生明显改变,祛瘀生新方可通过调节粪便中胆汁酸谱而减轻肠道炎症及症状.
Effect of decoction for dispeling stasis and generating new tissue on fecal bile acid metabolism in mice with mild to moderate ulcerative colitis
Objective It is to observe the effect of decoction for dispeling stasis and generating new tissue on bile acid metabolism in mice with mild to moderate ulcerative colitis(UC),and to explore its mechanism of action in treating UC.Methods Twenty-two cases C57BL/6J mice were taken,in which 5 mice were randomly selected as blank group,and the remaining rats were fed with 4%sodium glucan sulfate solution by free drinking to establish mild to moderate UC.The suc-cessfully modeled mice were randomly divided into model group,group of decoction for dispeling stasis and generating new tissue and mesalazine group,with 5 mice in each group.The group of decoction for dispeling stasis and generating new tis-sue and mesalazine group were given the corresponding drugs by gavage,and the blank group and model group were given distilled water by gavage,all once daily,continuously treated for 14 days.The activity,diet and defecation of mice in each group were observed;the fecal bile acid metabolism of mice was analyzed by targeted metabolomics technology at the end of the last gavage,and the histopathological morphology of colon was observed by HE staining.Results The mice in the model group were depressed,with lost weight,reduced hair,bloody and loose stools,and infiltration of a large number of inflam-matory cells and neutrophils in colonic tissues.The mice in the group of decoction for dispeling stasis and generating new tissue and mesalazine group were active at the end of gavage,their hair,diet and defecation were normal,with a few in-flammatory cells and neutrophils infiltrating in colonic tissue.Compared with the blank group,the contents of ursodeoxy-cholic acid(UDCA),lithocholic acid(LCA),tauro-α-mouse cholic acid(T-α-MCA),taurohyocholate(THCA),glyco-hyocholic acid(GHCA),ursocholic acid(UCA),hyocholic acid(HCA),deoxycholic acid(DCA),α-mouse cholic acid(α-MCA),taurochenodeoxycholic acid(TCDCA),taurodeoxycholic acid(TDCA),glycosaminoglutaric deoxycholic acid(GHDCA),hyodeoxycholic acid(HDCA),glycosaminolithocholic acid(GLCA),taurosulfocholic acid(TCA),and cho-lic acid(CA)were significantly decreased(all P<0.05),the contents of chenodeoxycholic acid(CDCA),β-mouse cho-lic acid(β-MCA),glycosaminolithocholic acid(GCA),dehydrocholic acid(dehydro-CA)were significantly increased in the model group(all P<0.05);compared with the model group,the contents of UDCA,LCA,T-α-MCA,THCA,GH-CA,UCA,α-MCA,HCA,DCA,TCDCA,TDCA,GHDCA,HDCA,GLCA,TCA and CA were significantly increased(all P<0.05),and the contents of CDCA,β-MCA,GCA,and dehydro-CA were significantly decreased in the group of decoction for dispeling stasis and generating new tissue and mesalazine group(all P<0.05).Conclusion The bile acid profile in the feces of mice with mild-to-moderate ulcerative colitis is significantly changed,and decoction for dispeling sta-sis and generating new tissue can alleviate intestinal inflammation and symptoms via regulating the bile acid profile in feces.
