Optimization of extraction process of Shenhua Decoction based on pharmacodynamics and its spectrum-effect relationship in the treatment of psoriasis
Objective It is to establish the HPLC fingerprint of Shenhua Decoction of different technological process and evaluate the spectrum-effect relationship in the treatment of psoriasis,so as to provide evidences for the development of new drugs for Shenhua Decoction.Methods The fingerprints of water extract,alcohol extract,and water extract with alco-hol precipitation of Shenhua Decoction were established by HPLC method,and their similarity among the samples was ana-lyzed.The psoriasis-like mouse models were established by imiquimod induction,and the therapeutic effects of different ex-tracts on the mouse skin injury were investigated.The scores of psoriasis area and severity index(PASI)and epidermal thickness of mouse skin lesions were used as pharmacodynamic indicators,and the spectrum-effect relationship between each common peak and anti-psoriasis effect was analyzed by grey correlation method.Results A total of 18 common peaks were identified in different extracts of Shenhua Decoction,and the correlation degree was not less than 0.922,and 4 char-acteristic peaks were identified(identified peak 1 was sophocarpidine,peak 5 was chlorogenic acid,peak 12 was galuteo-lin,and peak 17 was buddleoglucoside).The three extracts could reduce the PASI score and epidermal thickness of the skin lesions in mice,and the efficacy of water extract was the best.Among the common peaks,there were 16 and 15 peaks with great correlation with psoriasis efficacy index PASI score and epidermal thickness,respectively.Conclusion A quality evaluation method for the fingerprint profile of Shenhua Decoction was established in this study,water extract is the optimal extraction process,and its anti-psoriasis effect is the result of the joint action of multiple components.
