愈痫灵方对戊四氮致痫大鼠Pannexin1炎症信号通路蛋白及炎症因子表达的影响

目的 探讨愈痫灵方对戊四氮致痫大鼠Pannexin1(PANX1)炎症信号通路及炎症因子表达的影响.方法 取健康雄性SD大鼠80 只,随机选择10 只作为对照组,腹腔注射生理盐水;其余70 只腹腔注射戊四氮建立癫痫模型.将造模成功的40 只大鼠随机分为模型组、愈痫灵方组、丙磺舒组和丙戊酸钠组,每组10 只.愈痫灵方组给予3.24 g/(kg·d)愈痫灵方灌胃,丙磺舒组给予13.3 mg/(kg·d)丙磺舒灌胃,丙戊酸钠组给予31.9 mg/(kg·d)丙戊酸钠灌胃,模型组和对照组给予同等体积蒸馏水灌胃,均1 次/d,连续灌胃 14 d.末次灌胃结束后取大鼠颞叶内侧海马,采用Western blot法检测海马组织中PANX1、白细胞介素-1β(IL-1β)、p65、磷酸化p65(p-p65)蛋白表达情况;取腹主动脉血,采用ELISA法检测血清IL-1β、白细胞介素-6(IL-6)、核因子κB(NF-κB)水平.结果 模型组海马组织中PANX1、IL-1β、p65、p-p65 蛋白相对表达量均明显高于对照组(P均<0.05);愈痫灵方组、丙戊酸钠组、丙磺舒组海马组织中PANX1 蛋白相对表达量均明显低于模型组(P均<0.05),且愈痫灵方组明显低于对照组及丙磺舒组(P均<0.05),愈痫灵方组与丙戊酸钠组比较差异无统计学意义(P>0.05);愈痫灵方组及丙戊酸钠组海马组织中IL-1β、p65、p-p65 蛋白相对表达量均明显低于模型组和丙磺舒组(P均<0.05),丙戊酸钠组均明显低于愈痫灵方组(P均<0.05).模型组血清IL-1β、IL-6、NF-κB水平均明显高于对照组(P均<0.05);愈痫灵方组、丙戊酸钠组血清IL-1β水平均明显低于模型组和丙磺舒组(P均<0.05),愈痫灵组与丙戊酸钠组比较差异无统计学意义(P>0.05);愈痫灵方组血清IL-6、NF-κB水平均明显高于丙戊酸钠组(P均<0.05),与模型组比较差异均无统计学意义(P均>0.05).结论 愈痫灵方可以降低癫痫大鼠海马组织中PANX1 炎症信号通路蛋白的表达,减少下游炎症因子IL-1β、p65、p-p65 的产生,这可能是其抗癫痫的作用机制之一.
Effects of Yuxianling decoction on expressions of Pannexin1 inflammatory signaling pathway proteins and inflammatory factors in rats with epilepsy caused by pentylenetetrazol
Objective It is to explore the effects of Yuxianling decoction on expressions of Pannexin1 inflammatory sig-naling pathway proteins and inflammatory factors in rats with epilepsy caused by pentylenetetrazol(PTZ).Methods Eighty healthy male SD rats were taken,in which 10 rats were randomly selected as control group and injected intraperitoneally with normal saline,the remaining 70 rats were injected intraperitoneally with PTZ to establish models of epilepsy.40 suc-cessfully modeled rats were randomly divided into model group,Yuxianling decoction group,probenecid group and sodium valproate group,with 10 rats in each group.The Yuxianling decoction group was given Yuxianling decoction3.24 g/(kg·d)by gavage,the probenecid group was given probenecid 13.3 mg/(kg·d)by gavage,the sodium valproate group was given sodium valproate 31.9 mg/(kg·d)by gavage,and the model group and control group were given the same volume of dis-tilled water by gavage,all once daily,continuously treated for 14 days.At the end of the final gavage,the medial hippo-campus of temporal lobe of the rats was taken to detect the protein expressions of PANX1,interleukin-1β(IL-1β),p65 and phosphorylated p65(p-p65)in hippocampal tissues by Western blot;the blood of abdominal aortic artery was taken to detect the serum levels of IL-1β,interleukin-6(IL-6),and nuclear factor-κB(NF-κB)by ELISA.Results The relative expressions of PANX1,IL-1β,p65 and p-p65 proteins in hippocampal tissues of the model group were significantly higher than those of the control group(all P<0.05);the relative expressions of PANX1 protein in hippocampal tissues of the Yuxianling decoction group,sodium valproate group and probenecid group were significantly lower than that of the model group(all P<0.05),and the Yuxianling decoction group was lower than the control group and probenecid group(both P<0.05),there was no significant difference between the Yuxianling decoction group and sodium valproate group(P>0.05);the relative expressions of IL-1β,p65 and p-p65 proteins in hippocampal tissues of the Yuxianling decoction group and sodium valproate group were significantly lower than those of the model group and probenecid group(all P<0.05),and the sodium valproate group was significantly lower than the Yuxianling decoction group(P<0.05).The serum levels of IL-1β,IL-6 and NF-κB in the model group were significantly higher than those in the control group(all P<0.05);the serum levels of IL-1β in the Yuxianling group and sodium valproate group were significantly lower than those in the model group and probenecid group(all P<0.05),and there was no statistical significance between the Yuxianling group and so-dium valproate group(P>0.05);the serum levels of IL-6 and NF-κB in the Yuxianling group were significantly higher than those in the sodium valproate group(all P<0.05),but the differences were not statistically significant compared with the model group(all P>0.05).Conclusion Yuxianling decoction can reduce the expression of PANX1 channel in hipp-ocampal tissues and decrease the production of downstream inflammatory factors IL-1β,IL-6,P65 and P-P65 in rats with epilepsy,which may be one of the mechanisms of its antiepileptic action.
