Effects of lycium barbarum polysaccharide on Th1/Th2 cytokines in allergic rhinitis mice via NF-κB signaling pathway
Objective It is to investigate the effect of lycium barbarum polysaccharide(LBP)on Th1/Th2 cytokines in allergic rhinitis mice by modulating NF-κB signaling pathway.Methods Seventy-two SPF-grade SD mice were randomly di-vided into normal group,model group,fluticasone propionate group,and low,medium,and high dose groups of LBP,with 12 mice in each group.The mice in all groups except for the normal group were sensitized by ovalbumin to establish models of allergic rhinitis.After successful modeling,the fluticasone propionate group was treated with fluticasone propionate by nasal drops every day,and the low,medium,and high dose groups of LBP were respectively treated with 25 mg/kg,50 mg/kg and 100 mg/kg of LBP by gastric gavage every day,and the normal group and model group were given the same vol-ume of saline by gavage at the same time,all continuously treated for 21 days.The nasal symptom score of mice in each group were compared,the histopathological morphology of the nasal mucosa was observed by HE staining,the serum levels of interferon-gamma(IFN-γ),interleukin-4(IL-4)and immunoglobulin E(IgE)were detected by ELISA,and the pro-tein expressions of Toll-like receptor 4(TLR4),nuclear factor-κB(NF-κB)p65,and phosphorylated NF-κB p65(p-NF-κB p65)in nasal mucous membranes of mice were detected by Western-Blot.Results Compared with the normal group,the nasal symptom score,the serum levels of IL-4,IgE and the relative protein expressions of TLR4 and p-NF-κB p65 in nasal mucosa tissues of mice in the model group were significantly increased(all P<0.05),the serum level of IFN-γ of mice in the model group was significantly decreased(P<0.05),with much eosinophilic infiltration,glandular hyperplasia in nasal mucosal tissue;compared with the model group,the nasal symptom score,the serum levels of IL-4,IgE and the relative protein expressions of TLR4 and p-NF-κB p65 in nasal mucosa tissues of mice in each treatment group were significantly decreased(all P<0.05),the serum levels of IFN-γ of mice in each treatment group were significantly increased(P<0.05),the inflammatory reaction and glandular hyperplasia of nasal mucosal tissues were significantly improved,and the improvements of every indexes were more obvious with the increase of dosage of LBP.Conclusion Lycium barbarum poly-saccharide can effectively inhibit the expression of TLR4/NF-κB signaling pathway-related proteins,and reduce the produc-tion of IgE by restoring the imbalance of T-lymphocytes,thus to effectively attenuated the inflammatory response of nasal mucosa in mice with allergic rhinitis.
