基于NF-κB信号通路探讨枸杞多糖对过敏性鼻炎小鼠Th1/Th2细胞因子的影响

目的 探讨枸杞多糖调控核因子-κB(NF-κB)信号通路对过敏性鼻炎小鼠Th1/Th2 细胞因子的影响.方法 将72 只SPF级BALB/c 小鼠随机分为为正常组、模型组、丙酸氟替卡松组及枸杞多糖低、中、高剂量组,每组12 只.除正常组外,其余各组小鼠均采用卵清蛋白致敏法构建过敏性鼻炎模型.建模成功后,丙酸氟替卡松组每天予以丙酸氟替卡松滴鼻,枸杞多糖低、中、高剂量组每天予以25 mg/kg、50 mg/kg、100 mg/kg的枸杞多糖灌胃,正常组与模型组予以等量生理盐水灌胃,均持续干预21 d.比较各组小鼠鼻部症状评分,HE染色观察鼻黏膜组织病理学形态,ELISA法检测血清干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)、免疫球蛋白E(IgE)水平,Western blot法检测鼻黏膜组织中Toll样受体4(TLR4)、NF-κB p65 和磷酸化NF-κB p65(p-NF-κB p65)蛋白表达情况.结果 与正常组比较,模型组小鼠鼻部症状评分明显升高(P<0.05);鼻黏膜组织中有较多嗜酸性粒细胞浸润,腺体增生;血清IL-4、IgE水平及鼻黏膜组织中TLR4、p-NF-κB p65 蛋白相对表达量均明显升高(P均<0.05),血清IFN-γ水平明显降低(P<0.05).与模型组比较,各给药组小鼠鼻部症状评分明显降低(P均<0.05);鼻黏膜组织炎性反应和腺体增生均明显减轻;血清IL-4、IgE水平及鼻黏膜组织中TLR4、p-NF-κB p65 蛋白相对表达量均明显降低(P均<0.05),血清IFN-γ水平均明显升高(P均<0.05).且随枸杞多糖剂量的增加,各指标改善越明显(P均<0.05).结论 枸杞多糖可以通过抑制TLR4/NF-κB信号通路相关蛋白的表达,纠正Th1/Th2 细胞因子失衡,减少IgE的生成,进而有效减轻过敏性鼻炎小鼠的鼻黏膜炎症反应.
Effects of lycium barbarum polysaccharide on Th1/Th2 cytokines in allergic rhinitis mice via NF-κB signaling pathway
Objective It is to investigate the effect of lycium barbarum polysaccharide(LBP)on Th1/Th2 cytokines in allergic rhinitis mice by modulating NF-κB signaling pathway.Methods Seventy-two SPF-grade SD mice were randomly di-vided into normal group,model group,fluticasone propionate group,and low,medium,and high dose groups of LBP,with 12 mice in each group.The mice in all groups except for the normal group were sensitized by ovalbumin to establish models of allergic rhinitis.After successful modeling,the fluticasone propionate group was treated with fluticasone propionate by nasal drops every day,and the low,medium,and high dose groups of LBP were respectively treated with 25 mg/kg,50 mg/kg and 100 mg/kg of LBP by gastric gavage every day,and the normal group and model group were given the same vol-ume of saline by gavage at the same time,all continuously treated for 21 days.The nasal symptom score of mice in each group were compared,the histopathological morphology of the nasal mucosa was observed by HE staining,the serum levels of interferon-gamma(IFN-γ),interleukin-4(IL-4)and immunoglobulin E(IgE)were detected by ELISA,and the pro-tein expressions of Toll-like receptor 4(TLR4),nuclear factor-κB(NF-κB)p65,and phosphorylated NF-κB p65(p-NF-κB p65)in nasal mucous membranes of mice were detected by Western-Blot.Results Compared with the normal group,the nasal symptom score,the serum levels of IL-4,IgE and the relative protein expressions of TLR4 and p-NF-κB p65 in nasal mucosa tissues of mice in the model group were significantly increased(all P<0.05),the serum level of IFN-γ of mice in the model group was significantly decreased(P<0.05),with much eosinophilic infiltration,glandular hyperplasia in nasal mucosal tissue;compared with the model group,the nasal symptom score,the serum levels of IL-4,IgE and the relative protein expressions of TLR4 and p-NF-κB p65 in nasal mucosa tissues of mice in each treatment group were significantly decreased(all P<0.05),the serum levels of IFN-γ of mice in each treatment group were significantly increased(P<0.05),the inflammatory reaction and glandular hyperplasia of nasal mucosal tissues were significantly improved,and the improvements of every indexes were more obvious with the increase of dosage of LBP.Conclusion Lycium barbarum poly-saccharide can effectively inhibit the expression of TLR4/NF-κB signaling pathway-related proteins,and reduce the produc-tion of IgE by restoring the imbalance of T-lymphocytes,thus to effectively attenuated the inflammatory response of nasal mucosa in mice with allergic rhinitis.
