壮药白花蛇舌草水提物对MDA-MB-231乳腺癌裸鼠移植瘤的抑制及免疫调节作用研究

目的 观察白花蛇舌草水提物对MDA-MB-231 乳腺癌裸鼠移植瘤的抑制及免疫调节作用,并探讨其作用机制.方法 取 6 只正常BALB/c-nu雌性裸鼠作为正常组;取 30 只BALB/c-nu雌性裸鼠复制MDA-MB-231 乳腺癌移植瘤模型,将造模成功裸鼠随机分为模型组、环磷酰胺组及白花蛇舌草水提物低、高剂量组,每组6 只.环磷酰胺组给予环磷酰胺25 mg/kg腹腔注射,每7d注射1 次;白花蛇舌草水提物低、高剂量组分别按照1 500 mg/(kg·d)、6 000 mg/(kg·d)灌胃白花蛇舌草水提物溶液,正常组和模型组灌胃等体积灭菌注射用水,均1 次/d.各组均连续干预21 d,监测裸鼠活动状况,每间隔2d测量1 次肿瘤大小.干预结束后,测量体重、瘤重与脾重,计算脾脏指数、抑瘤率;HE染色观察移植瘤的病理学形态;脾淋巴细胞增殖试验评价脾脏中T淋巴细胞和B淋巴细胞的增殖能力;血液分析仪测定外周血中白细胞数目和淋巴细胞占比;ELISA法检测血清中白细胞介素-2(IL-2)、白细胞介素-6(IL-6)、γ干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)水平;Western blot法测定模型组和白花蛇舌草水提物低、高剂量组裸鼠肿瘤组织中磷脂酰肌醇3-激酶(PI3K)/蛋白激B(Akt)/哺乳动物雷帕霉素靶蛋白(mTOR)通路蛋白及凋亡相关蛋白[B细胞淋巴瘤/白血病-2 蛋白(Bcl-2)、Bcl-2 相关X蛋白(Bax)、半胱天冬氨酸蛋白酶-7(Caspase-7)、半胱天冬氨酸蛋白酶-9(Caspase-9)]表达情况.结果 与正常组比较,模型组裸鼠脾脏指数、外周血中白细胞数量均明显升高(P均<0.05),脾脏T淋巴细胞和B淋巴细胞增殖能力指数、外周血淋巴细胞占比及血清中IL-2、IL-6、IFN-γ、TNF-α水平均明显降低(P均<0.05);肿瘤组织中肿瘤细胞密集丰富排列无序,肿瘤细胞异型性明显.与模型组比较,白花蛇舌草水提物高、低剂量组裸鼠脾脏指数、脾脏T淋巴细胞和B 淋巴细胞增殖能力指数、外周血淋巴细胞占比及血清中IL-2、IL-6、IFN-γ、TNF-α水平均明显升高(P均<0.05),瘤重、外周血中白细胞数量均明显降低(P均<0.05);肿瘤组织中肿瘤细胞数目大量减少,细胞凋亡,组织结构破损;肿瘤组织中p-PI3K、PI3K、p-Akt、Akt、mTOR、Bcl-2 蛋白相对表达量均明显降低(P均<0.05),Bax、Caspase-7、Caspase-9 蛋白相对表达量均明显升高(P均<0.05).环磷酰胺组裸鼠脾脏指数、瘤重、脾脏T淋巴细胞和B 淋巴细胞增殖能力指数、外周血白细胞数量和淋巴细胞占比均明显低于模型组(P均<0.05);肿瘤组织中肿瘤细胞数量明显减少.结论 白花蛇舌草水提物可显著抑制MDA-MB-231 乳腺癌裸鼠移植瘤生长,诱导乳腺癌细胞发生凋亡,增强移植瘤裸鼠免疫功能,其机制可能与抑制PI3K/Akt/mTOR通路激活,下调凋亡相关蛋白表达、上调促凋亡蛋白表达相关.
Anti-tumor and immunomodulatory effects of Hedyotis diffusa Willd.water extract on human breast cancer MDA-MB-231 transplanted tumor in nude mice
Objective It is to investigate the anti-tumor and immunomodulatory effects of Hedyotis diffusa Willd.water extract(HDWE)on human breast cancer MDA-MB-231 xenograft tumor in nude mice,and to explore its mechanism.Methods Six normal BALB/c-nu female nude mice were selected as normal group,another 30 BALB/c-nu female nude mice were used to prepare models of MDA-MB-231 breast cancer transplanted tumor.The successfully modeled mice were randomly divided into model group,cyclophosphamide(CTX)group and HDWE low-dose group and HDWE high-dose group,with 6 mice in each group.The CTX group was given CTX 25 mg/kg by intraperitoneal injection every 7 days,the low and high dose groups of HDWE were gavaged with HDWE 1 500 mg/(kg·d)and 6 000 mg/(kg·d)respectively,and the normal and model groups were gavaged with equal volume of sterilized injectable water,all once daily,continuously treated for 21 days.The activity status of the nude mice was monitored,and their tumor sizes were measured once every 2 days interval.At the end of the treatment,the body weight,tumor weight and spleen weight were measured,the spleen in-dex and tumor suppression rate were calculated;the pathological morphology of the transplanted tumors was observed by HE staining;the proliferative capacity of T and B lymphocytes in spleen was evaluated by splenic lymphocyte proliferation as-say;the number of leukocytes and percentage of lymphocytes in peripheral blood were detected by hematology analyzer;the serum levels of IL-2,IL-6,IFN-γ and TNF-α were detected by ELISA;the expressions of phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR)pathway proteins and apoptosis-related proteins(Bcl-2,Bax,Caspase-7 and Caspase-9)in tumor tissues of nude mice of the model group and low and high dose groups of HDWE were detected by Western blot assay.Results Compared with the normal group,the spleen index and number of leukocytes in peripheral blood were significantly increased,while the proliferative capacity of T and B lymphocytes in spleen,percentage of lymphocytes in peripheral blood,serum levels of IL-2,IL-6,IFN-γ and TNF-α were significantly de-creased in the nude mice of the model group(all P<0.05),the tumor cells in tumor tissue were densely abundantly ar-ranged in disorder,and their heterogeneity was obvious.Compared with the model group,the spleen index,proliferative capacity of T and B lymphocytes in spleen,percentage of lymphocytes in peripheral blood,and serum levels of IL-2,IL-6,IFN-γ,and TNF-α were significantly increased,while the tumor weight and number of leukocytes in peripheral blood were all significantly reduced in the nude mice of the the low and high dose groups of HDWE(all P<0.05);the number of tumor cells in tumor tissue was greatly reduced,with apoptosis and structural damage;the relative expressions of p-PI3K,PI3K,p-Akt,Akt,mTOR,and Bcl-2 proteins in tumor tissues were significantly reduced(all P<0.05),and the relative expressions of Bax,Caspase-7,and Caspase-9 proteins were significantly increased(all P<0.05).The spleen index,tumor weight,the proliferative capacity of T and B lymphocytes in spleen,the number of leukocytes and percentage of lym-phocytes in peripheral blood in the nude mice of the CTX group were significantly lower than those in the model group(all P<0.05),and the number of tumor cells in tumor tissue was significantly reduced.Conclusion HDWE can significantly inhibit the growth of transplanted tumors,induce apoptosis of breast cancer cells,and enhance immune function of nude mice with MDA-MB-231 breast cancer,the mechanism of which may be related to inhibit the activation of PI3K/Akt/mTOR pathway,down-regulate the expressions of apoptosis-associated proteins,and up-regulate the expressions of pro-ap-optotic proteins.
