盆炎通络方对输卵管炎性不孕大鼠血清炎症因子的影响

目的 观察盆炎通络方对输卵管炎性不孕大鼠血清炎症因子及输卵管组织结构的影响,探讨盆炎通络方治疗输卵管炎性不孕的可能作用机制.方法 将90 只SD雌性大鼠随机分为空白组、模型组、康妇炎胶囊组及盆炎通络方低、中、高剂量组,每组15 只.除空白组外,其余组大鼠均采用双侧输卵管注射苯酚糊剂方法建立输卵管炎性不孕模型.造模成功后,康妇炎胶囊组给予康妇炎混悬液0.016 g/mL灌胃,盆炎通络方低、中、高剂量组分别给予盆炎通络方混悬液0.55 g/mL、1.1 g/mL、2.2 g/mL灌胃,空白组及模型组予以蒸馏水灌胃,均2 次/d,连续灌胃4 周.末次灌胃结束后,ELISA法检测血清肿瘤坏死因子-α(TNF-α)、γ干扰素(IFN-γ)、白细胞介素-1(IL-1)、白细胞介素-2(IL-2)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)水平,肉眼观察输卵管外观形态.结果 模型组大鼠血清TNF-α、IFN-γ、IL-1、IL-2、IL-6 水平均明显高于空白组(P均<0.05),血清IL-10 水平明显低于空白组(P<0.05);盆炎通络方中、高剂量组和康妇炎胶囊组血清TNF-α、IFN-γ、IL-1、IL-2、IL-6 水平均明显低于模型组(P均<0.05),血清IL-10 水平均明显高于模型组(P均<0.05);盆炎通络方中、高剂量组血清TNF-α、IFN-γ、IL-1、IL-2、IL-6 水平均明显低于盆炎通络方低剂量组(P均<0.05),血清IL-10 水平均明显高于盆炎通络方低剂量组(P均<0.05),盆炎通络方中、高剂量组上述血清炎症因子水平比较差异均无统计学意义(P均>0.05).模型组大鼠输卵管组织肿胀明显,色苍白无光泽,与周围组织形成致密粘连,可见积水;盆炎通络方低剂量组外观形态与模型组相近;盆炎通络方中、高剂量组和康妇炎胶囊组输卵管组织轻微增厚,色红有光泽,与周围组织无明显粘连或有少许粘连,无积水,其中盆炎通络方中剂量组更接近于正常.结论 盆炎通络方可通过降低血清TNF-α、IFN-γ、IL-1、IL-2、IL-6 水平,升高IL-10 水平,从而有效改善输卵管炎性不孕大鼠的输卵管炎性病变.
Effect of decoction for unblocking collaterals for pelvic inflammation on serum inflammatory factors in rats with tubal inflammatory infertility
Objective It is to observe the effects of decoction for unblocking collaterals for pelvic inflammation on ser-um inflammatory factors and tubal tissue structure in rats with tubal inflammatory infertility,and to explore the possible mechanism of action of the decoction in the treatment of tubal inflammatory infertility.Methods Ninety SD female rats were randomly divided into blank group,model group,Kangfuyan capsule group and low,medium and high dose groups of de-coction for unblocking collaterals for pelvic inflammation,with 15 rats in each group.The rats of all group except for the blank group were given phenol paste by bilateral tubal injection to establish models of tubal inflammatory infertility.After successful modeling,the Kangfuyan capsule group was given Kangfuyan suspension 0.016 g/mL by gavage,the low,medi-um and high dose groups of decoction for unblocking collaterals for pelvic inflammation were given suspension of decoction for unblocking collaterals for pelvic inflammation 0.55 g/mL,1.1 g/mL and 2.2 g/mL by gavage,respectively,and the blank group and model group were given distilled water by gavage,all twice daily,continuously treated for 4 weeks.At the end of the final gavage,the serum levels of tumor necrosis factor-α(TNF-α),gamma interferon(IFN-γ),interleukin-1(IL-1),interleukin-2(IL-2),interleukin-6(IL-6)and interleukin-10(IL-10)were detected by ELISA,and the appear-ance and morphology of the fallopian tubes were observed by naked eyes.Results The serum levels of TNF-α,IFN-γ,IL-1,IL-2 and IL-6 of rats in the model group were significantly higher,while the serum level of IL-10 was significantly lower than those in the blank group(all P<0.05);the serum levels of TNF-α,IFN-γ,IL-1,IL-2 and IL-6 levels were signifi-cantly lower,while the serum levels of IL-10 levels were significantly higher in medium-dose and high-dose groups of de-coction for unblocking collaterals for pelvic inflammation and Kangfuyan capsule group than those in the model group(all P<0.05);the serum levels of TNF-α,IFN-γ,IL-1,IL-2 and IL-6 levels were significantly lower,while the serum levels of IL-10 levels were significantly higher in medium-dose and high-dose groups of decoction for unblocking collaterals for pelvic inflammation than those in the low dose group(all P<0.05),there was no significant difference in the levels of the above serum inflammatory factors between the high-dose group and medium dose group(P>0.05).In the rats of the model group,the oviduct tissue was obviously swelling,pale and lusterless,formed dense adhesions with surrounding tissues,and with fluid retention;the appearance and morphology of the low dose group of decoction for unblocking collaterals for pelvic inflammation were similar to those of the model group;the oviduct tissue of the medium dose and high dose groups of decoc-tion for unblocking collaterals for pelvic inflammation and Kangfuyan capsule group were slightly thickened,reddish and shiny,with no obvious adhesion with the surrounding tissues or with a little adhesion,and with no water retention,of which the medium dose group was closer to normal.Conclusion Decoction for unblocking collaterals for pelvic inflammation can effectively improve tubal inflammatory lesions in rats with tubal inflammatory infertility via reducing the serum levels of TNF-α,IFN-γ,IL-1,IL-2,IL-6,and increasing the serum level of IL-10.
