清化饮对湿热型抑郁大鼠血清多巴胺及小肠病理改变的影响

目的 探讨清化饮对湿热型抑郁大鼠血清多巴胺及小肠病理形态的影响.方法 取30只SD雄性大鼠,随机选择5只作为空白组,其余25只大鼠采用高脂高糖饮食与慢性不可预见性温和应激方法建立脾胃湿热型抑郁模型.将造模成功的25只大鼠随机分为模型组6只、清化饮组6只、舍曲林组6只、清化饮加舍曲林组7只,清化饮组给予清化饮27.5 g/(kg·d)灌胃,舍曲林组给予舍曲林片5.21 mg/(kg·d)灌胃,清化饮加舍曲林组予清化饮27.5 g/(kg·d)和舍曲林片5.21 mg/(kg·d)灌胃,空白组和模型组灌胃等容积的生理盐水,均1次/d,连续21 d.末次灌胃结束后进行行为学旷场实验,实验后取大鼠血清检测多巴胺水平,取小肠组织进行HE染色病理观察.结果 清化饮组、清化饮加舍曲林组大鼠中心穿格次数均明显多于模型组(P均<0.05),各药物组间大鼠中心穿格次数差异无统计学意义(P>0.05);清化饮加舍曲林组外围穿格次数均明显少于空白组和清化饮组(P均<0.05);清化饮组大鼠外围穿格次数有多于模型组、舍曲林组的趋势,接近于空白组,但4组间比较差异均无统计学意义(P均>0.05);各组大鼠的直立次数、理毛次数比较差异均无统计学意义(P均>0.05).模型组血清多巴胺水平与空白组比较差异无统计学意义(P>0.05),各药物组血清多巴胺水平均明显高于模型组(P均<0.05),舍曲林组、清化饮加舍曲林组血清多巴胺水平均明显高于空白组(P均<0.05).与空白组比较,模型组大鼠小肠上皮及固有膜内可见炎细胞,固有膜内淋巴小结形成;各药物组小肠上皮及固有膜内炎细胞较模型组有所减少,且清化饮组改善较其他2组更明显.结论 清化饮可改善湿热型抑郁大鼠的抑郁症状,提高大鼠血清多巴胺水平,对小肠黏膜有一定保护作用.
Effect of decoction for clearing and resolving on serum dopamine and small intestinal pathological changes in rats with depression of type of dampness and heat
Objective It is to explore the effects of decoction for clearing and resolving on serum dopamine and small intestinal pathology in rats with depression of type of dampness and heat.Methods Thirty SD male rats were taken,in which 5 rats were randomly selected as the blank group,and the remaining 25 rats were fed with high-fat and high-sugar di-et and stimulated with chronic unpredictable mild stress to establish models of spleen-stomach dampness-heat depression.The successfully modeled 25 rats were randomly divided into a model group of 6 rats,a decoction for clearing and resolving group of 6 rats,a sertraline group of 6 rats,and a decoction for clearing and resolving plus sertraline group of 7 rats.The decoction for clearing and resolving group was given decoction for clearing and resolving 27.5 g/(kg·d)by gavage,the sertraline group was given sertraline tablets 5.21 mg/(kg·d)by gavage,and the decoction for clearing and resolving plus sertraline group was given decoction for clearing and resolving 27.5 g/(kg·d)and sertraline tablets 5.21 mg/(kg·d)by gavage,the blank group and model group were given equal volumes of normal saline by gavage,all once daily,continuously traeted for 21 days.The actions of the rats were detected by open field test at the end of the final gavage,and after the test,the serum of the rats was taken to detect the level of dopamine,and their small intestinal tissues were taken to observe the pathology by HE staining.Results The times of crossing the center of the rats of decoction for clearing and resolving group and decoction for clearing and resolving plus sertraline group were significantly more than that of the model group(both P<0.05),but there was no significant difference in the times of crossing the center among each treatment group(all P>0.05);the times of crossing the peripheral of the decoction for clearing and resolving plus sertraline group were significant-ly less than those of the blank group and decoction for clearing and resolving group(both P<0.05);the times of crossing the peripheral of rats of the decoction for clearing and resolving group had a tendency to be more than that of the model group and sertraline group,and was close to that of the blank group,but the differences were not statistically significant when comparing among the 4 groups(all P>0.05);there was no significant difference in the times of upstanding and grooming of rats among each group(all P>0.05).There was no significant difference in the serum level of dopamine be-tween the model group and blank group(P>0.05),the serum level of dopamine of each treatment group was significantly higher than that of the model group(all P<0.05),and the serum level of dopamine of the sertraline group and decoction for clearing and resolving plus sertraline group were significantly higher than that of the blank group(both P<0.05).Compared with the blank group,there were inflammatory cells in epithelium and lamina propria of the small intestine with lymph node formation in the lamina propria in rats of the model group,the inflammatory cells in epithelium and lamina pro-pria of the small intestine were reduced in each treatment group compared with the model group,and the improvement was more significant in the decoction for clearing and resolving group compared with other two groups.Conclusion Decoction for clearing and resolving group can improve depressive symptoms,increase serum dopamine concentration in rats with depres-sion of type of dampness and heat,it can protect the small intestine mucosa.
