小鼠白癜风疾病模型的构建及初步研究

目的 比较氢醌乳膏和莫诺苯宗乳膏诱导野生型C57BL/6J小鼠产生白癜风样表型的作用,构建组织病理学相似度高、脱色效果持久稳定且成本低、易操作的白癜风样小鼠模型,为选择白癜风疾病动物模型以研究人类白癜风疾病的发病机制以及药物治疗效果、药物作用机制提供依据.方法 将30只C57BL/6J小鼠随机分为3组,每组10只.3组小鼠于背部相同部位去毛后,对照组涂抹乳膏基质,莫诺苯宗组涂抹40%莫诺苯宗乳膏,氢醌组涂抹2.5%氢醌乳膏,均1次/d,建模周期为50d.肉眼观察造模部位皮肤变化;建模结束后处死各组小鼠,取病变部位皮肤组织,进行酪氨酸酶(TYR)免疫荧光染色观察;摘眼球取血,ELISA法检测血清中TYR、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-9(MMP-9)水平.结果 莫诺苯宗组和氢醌组小鼠均出现明显皮肤脱色,TYR表达平均光密度值明显降低.莫诺苯宗组和氢醌组小鼠血清TYR水平均明显低于对照组(P均<0.05),莫诺苯宗组和氢醌组比较差异无统计学意义(P>0.05);氢醌组小鼠血清IFN-γ、MMP-9水平均明显高于对照组和莫诺苯宗组(P均<0.05),血清TNF-α水平与对照组比较差异无统计学意义(P>0.05);莫诺苯宗组小鼠血清TNF-α水平明显高于对照组(P均<0.05),血清IFN-γ、MMP-9水平与对照组比较差异均无统计学意义(P均>0.05).结论 2.5%氢醌乳膏和40%莫诺苯宗乳膏涂抹法均可成功建立野生型C57BL/6J小鼠白癜风模型,其中2.5%氢醌乳膏更易诱导建立白癜风表观模型.
Establishment and preliminary identification of models of mouse vitiligo disease
Objective It is to compare the effects of hydroquinone cream and monobenzone cream in inducing vitiligo like phenotype in wild-type C57BL/6J mice,and to construct cost-effective and easy to operate vitiligo like mouse models with high histopathological similarity and long-lasting decolorization effect.This provides a basis for selecting vitiligo like animal models for subsequent research on the pathogenesis of human vitiligo disease,as well as the therapeutic effect and drug action mechanism of drugs on this disease.Methods Thirty C57BL/6J mice were randomly divided into 3 groups,with 10 mice in each group.After the mice of the three groups were depilated on the same parts of back,the control group was applied with cream base,the monobenzone group was applied with 40%monobenzone cream,and the hydroquinone group was applied with 2.5%hydroquinone cream,all once daily,modeled for 50 days.The skin changes at the modeling site were observed with naked eyes;at the end of modeling,the mice in each group were executed,and their skin tissues were taken from the lesion site to detect the expression of tyrosinase(TYR)by immunofluorescence staining;the blood was col-lected from their eyeballs to determine the serum levels of TYR,interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α),and matrix metalloproteinase-9(MMP-9)by ELISA.Results The mice in both the monobenzone group and hydroquinone group had significant skin discoloration,and their mean optical density value of TYR expression was significantly reduced.The serum levels of TYR of mice in the monobenzone group and hydroquinone group were significantly lower than that in the control group(both P<0.05),and the difference between the monobenzone group and hydroquinone group was not statisti-cally significant(P>0.05);the serum levels of IFN-γ and MMP-9 of mice in the hydroquinone group were significantly higher than those of mice in the control group and monobenzone group(all P<0.05),and the difference in the serum level of TNF-α was not statistically significant between the monobenzone group and control group(P>0.05);the serum level of TNF-α in the monobenzone group was significantly higher than those in the control group and hydroquinone group(both P<0.05),and the differences in serum levels of IFN-γ and MMP-9 were not statistically significant when compared with the control group(both P>0.05).Conclusion Both 2.5%hydroquinone cream and 40%monobenzone cream application methods can successfully establish vitiligo models in wild-type C57BL/6J mice,2.5%hydroquinone cream is more easily to induce vitiligo phenotype.
