Expression of fibronectin 2 in ovarian cancer and its impact on the biological characteristics of SKOV3 cells
Objective It is to explore the expression of fibronectin 2(FGL2)in epithelial ovarian cancer,and to study its effect on the biological characteristics of human ovarian cancer SKOV3 cells and correlated mechanisms.Methods ①The resected tissue specimens were taken from 40 cases of patients with serous ovarian carcinoma and 16 cases of patients with serous cystadenoma of ovary who underwent surgical treatment in Hebei General Hospital,and the expression of FGL2 protein in the tissue specimens was detected by immunohistochemistry.②The SKOV3 cells were taken and divided into FGL2 siRNA transfection group and negative control siRNA group,the FGL2 protein expression in the cells was detected by Western blot to determine the efficiency of FGL2 siRNA transfection,then the proliferation,invasion and migration ability of SKOV3 cells in the FGL2 siRNA transfection group and negative control siRNA group were subsequently detected by CCK-8 assay and Transwell assay,and the protein expressions of interleukin-6(IL-6),signal transducer and activator of transcription 3(STAT3)in the cells were detected by Western blot assay,so as to observe the effect of silencing of FGL2 on the proliferation,invasion and migration ability of SKOV3 cells and on IL-6/STAT3 signaling pathway.Results The high expression rate of FGL2 protein in tissue specimens from patients with serous ovarian carcinoma was significantly higher than that from patients with serous cystadenoma of ovary[80.0%(32/40)versus 37.5%(6/16),P<0.05].The rela-tive expression of FGL2 protein in the FGL2 siRNA-transfection group was significantly lower than that of the negative con-trol siRNA group(P<0.05);the proliferative activity of SKOV3 cells at 48 h,72 h,96 h and 120 h after transfection in the FGL2 siRNA-transfection group was significantly lower than that of the negative control siRNA group(P<0.05);Tran-swell invasion assay with matrix gel and migration assay without matrix gel showed that the number of membrane-penetrating cells in the FGL2 siRNA transfection group was significantly less than that in the negative control siRNA group(P<0.05);the relative expression of 1L-6 and p-STAT3 proteins in the FGL2 siRNA group were both significantly lower than those in the negative control siRNA group(both P<0.05).Conclusion There is high FGL2 expression in ovarian cancer tissues,silencing FGL2 to inhibit the activity of IL-6/STAT3 signaling pathway can inflence the biological characteristics of ovarian epithelial cancer cells,which suggests that FGL2 may serve as one of the potential targets for the diagnosis and treatment of ovarian cancer.
