Protective effect and mechanism of Jinshuibao on osteoporosis in ovariectomized mice
Objective It is to investigate the protective effect and mechanism of Jinshuibao on osteoporosis in ovariecto-mized mice.Methods Thirty female C57BL/6J mice were randomly divided into control group,model group and Jinshuibao group,with 10 mice in each group.The mice of model group and Jinshuibao group were treated with ovariectomy to estab-lish models of postmenopausal osteoporosis.Starting from the 10th day of modeling,the Jinshuibao group was given Jin-shuibao suspension 386 mg/kg by gavage,while the control group and model group were given equal volumes of double dis-tilled water by gavage,all once daily,for 6 consecutive days per week,and continuously gavaged for 15 weeks.The femur bone microstructure(percent bone volume,trabecular number,trabecular separation)and bone mineral density of the mice was detected by Micro-CT,the serum levels of C-terminal telopeptide of type Ⅰ collagen(CTX-Ⅰ),N-terminal propeptide of type Ⅰ procollagen(PINP),bone alkaline phosphatase(BALP)and tartrate-resistant acid phosphatase(TRAP)of the mice were measured by ELISA,and the protein expressions of hypoxia-inducible factor-2α(HIF-2α)and tumor necrosis factor receptor associated factor 6(TRAF6)in bone tissue of the mice were detected by Western blot.Results Compared with the control group,the femur bone microstructure of the mice of the model group was disrupted,and the percent bone volume,trabecular number and bone mineral density were significantly reduced(all P<0.05),the trabecular separation,serum levels of CTX-Ⅰ,PINP,BALP,TRAP,and relative protein expressions of HIF-2α and TRAF6 in bone tissue were significantly increased(all P<0.05).Compared with the model group,the damage of the femur bone microstructure of the mice of Jinshuibao group was significantly improved,and the percent bone volume,trabecular number and bone mineral density were significantly increased(all P<0.05),the trabecular separation,serum levels of CTX-Ⅰ,PINP,BALP,TRAP,and relative protein expressions of HIF-2α and TRAF6 in bone tissue were significantly decreased(all P<0.05).Conclusion Jinshuibao can effectively improve osteoporosis in ovariectomized mice,and the mechanism may be related to inhibiting the HIF-2α/TRAF6 signaling pathway and regulating bone metabolism.
