Effects of tanshinone Ⅱ A on testicular torsion-induced ischemia-reperfusion injury in adult rats
Objective It is to observe the protective effect of tanshinone Ⅱ A on ischemia-reperfusion(IR)injury after testicular torsion/detorsion in adult rats.Methods Sixty-four male SD rats were randomly divided into sham operation group,model group,tanshinone Ⅱ A single and continuous treatment groups,with 16 rats in each group.The models of IR after testicular torsion/detorsion were established in the rats of all groups except for the sham operation group.The single treatment group was injected intraperitoneally with tanshinone Ⅱ A 25 mg/kg after torsion/detorsion,while the continuous treatment group was maintained with tanshinone Ⅱ A 25 mg/kg once per day for one week in addition to the above proce-dures.Eight rats were randomly taken and executed from each group on the 4th and 29th postoperative days,respectively.The histopathological morphology of testicular tissue and the apoptosis of testicular spermatogonial cells on the 4th day after surgery were observed by HE staining and TUNEL fluorescence staining,and the contents of malondialdehyde(MDA),su-peroxide dismutase(SOD),glutathione peroxidase(GSH-Px)were determined by thiobarbituric acid method,hydroxyla-mine method and ultraviolet colorimetric method,the protein expressions of Bcl-2 and Bax in testicular tissue on the 29th day after surgery were detected by Western blot method.Results On the 4th day after surgery,the testicular spermatogenic cells of rats in the model group were loosely and disorganized arranged,some spermatogenic cells were detached,the mes-enchyme was edematous and congested,and spermatogenic cells were more apoptotic,the spermatogenic cell apoptotic in-dex and the content of MDA in the testicular tissues were significantly higher while the contents of SOD and GSH-Px in the testicular tissues were significantly lower than those of the sham operation group(all P<0.05);compared with the model group,the spermatogenic cells of rats in the tanshinone Ⅱ A continuous treatment group were relatively tightly and orderly arranged,a few spermatogenic cells were detached,no obvious change of mesenchyme was seen,and spermatogenic cell was less apoptotic,the apoptotic index of spermatogenic cells and the content of MDA in the testicular tissues were signifi-cantly decreased(all P<0.05),and the contents of SOD and GSH-Px in the testicular tissues were significantly increased(P all<0.05),and the content of MDA was significantly lower,while the contents of SOD and GSH-Px in the testicular tissues were significantly higher in the tanshinone Ⅱ A continuous treatment group than those in the tanshinone Ⅱ A single treatment group(all P<0.05).On the 29th day after surgery,the relative protein expression of Bcl-2 in the testicular tis-sues of rats in the model group was significantly lower than that in the sham operation group(P<0.05),and the relative protein expression of Bax was significantly higher than that in the sham operation group(P<0.05);the relative protein ex-pression of Bcl-2 in the testicular tissues of rats in the tanshinone Ⅱ A continuous treatment group was significantly higher,while the relative protein expression of Bax was significantly lower than those in the model group and tanshinone Ⅱ A single treatment group(all P<0.05).Conclusion Continuous application of tanshinone Ⅱ A can attenuate oxidative stress after testicular ischemia-reperfusion injury in rats,and may reduce testicular spermatogenic cell apoptosis by up-regulating Bcl-2 protein expression and down-regulating Bax protein expression.
