Decoction for cultivating primordium and resisting cancer inhibits tumor growth in hepatocellular carcinoma mice by regulating autophagy through circ_SLIT3/NF-KB axis
Objective It is to observe the inhibition of decoction for cultivating primordium and resisting cancer on tumor growth in Huh7 hepatocellular carcinoma mice and its effects on circRNA slit guidance ligand 3(circ_SLIT3)/nucle-ar factor-KB(NF-κB)axis and autophagy of tumor cells.Methods Twelve male C57BL/6 mice were selected to establish models of hepatocellular carcinoma by subcutaneous inoculation of hepatocellular carcinoma Huh7 cell suspension in the right axilla.The successfully modeled mice were randomly divided into model group and group of decoction for cultivating primordium and resisting cancer,and were given normal saline and 2 g/mL decoction for cultivating primordium and resis-ting cancer by gavage,respectively.After 11 d of continuous intervention,the mice were executed,the tumor volume was calculated according to the length and width of the tumor,the tumor tissue was peeled off and weighed and the tumor inhibi-tion rate was calculated,the formation of autophagic vesicles and autophagic vesicles was observed by transmission electron microscopy,the expressions of microtubule-associated protein 1 light chain 3(LC3),Caspase-3,Bcl-2,Bax in tumor tis-sues were detected by Western blot method,and the expression of circ_SLIT3 mRNA in tumor tissues was detected by PCR.Results Compared with the model group,the general conditions such as mental status,eating and drinking status,etc.of mice in the group of decoction for cultivating primordium and resisting cancer were better,and the tumor volume and weight of the mice were significantly reduced(all P<0.05),cytolysosome and autophagic vesicles in the tumor cells were significantly increased;the ratio of LC3 Ⅱ/LC3 Ⅰ and the relative protein expressions of Caspase-3,Bax and NF-κB in the tumor tissues were all significantly increased(all P<0.05),the relative expressions of Bcl-2 protein and circ_SLIT3 mR-NA were all significantly reduced(all P<0.05).Conclusion Decoction for cultivating primordium and resisting cancer can inhibit tumor growth in Huh7 hepatocellular carcinoma mice,which may be achieved by regulating autophagy through circ_SLIT3/NF-κB axis.
