Effects of magnolol on the expression of IL-6 and VEGF in periodontal tissues of rats with periodontitis
Objective It is to observe the effects of magnolol on periodontal lesions and the expressions of interleukin-6(IL-6)and vascular endothelial growth factor(VEGF)in periodontal tissues of periodontitis rats,and to explore the val-ue of magnolol in the treatment of periodontitis.Methods Sixty-five 3-month-old male SD rats were selected,in which 10 rats were randomly selected as the control group,and the remaining rats were used to establish periodontitis models by liga-ting bilateral maxillary first molar teeth with orthodontic ligature wire.After 4 weeks of ligation,40 successfully modeled rats were randomly divided into 4 groups,with 10 rats in each group.Topical irritant ligature wire was removed in the topi-cal treatment group;the magnolol treatment group was given 50 mg/(kg·d)of magnolol by gavage without removing the ligature wire,the topical+magnolol treatment group was given 50 mg/(kg·d)of magnolol by gavage based on removing the ligature wire,and the control group was given an equal volume of saline by gavage,all the groups were treated for 8 weeks.The general condition of the periodontal tissues and dentition of the rats in each group were observed,the pathologi-cal morphology of the periodontal tissues was observed by HE staining,and the positive expressions of IL-6 and VEGF in the periodontal tissues were detected by immunohistochemical staining.Results The bilateral maxillary first molar gingiva of rats in the periodontitis group were red and swollen;the combined epithelium left the enamel-osseous border and shifted to-ward the root to form deep periodontal pockets surrounded by a large number of inflammatory cells infiltrated,with resorp-tion and destruction of the tops of alveolar ridges and intrinsic alveolar bone.The bilateral maxillary first molar gingiva of rats in the topical treatment group was red and swollen,with probing bleeding;the periodontal pockets were deep,with a small amount of inflammatory cell infiltration around them,and there was severe destruction of the alveolar ridge apex and intrinsic alveolar bone,with neoplastic bone formation on the alveolar ridge apex.The bilateral maxillary first molar gingiva of rats in the magnolol treatment group and the topical+magnolol treatment group was slight red and swollen or not red and swollen;the periodontal pockets were shallow,with a small amount of inflammatory cell infiltration around them and neo-plastic bone formation on the alveolar ridge apex,the destruction of the periodontal tissues in the topical+magnolol treat-ment group was more mild.The mean optical densities of positive expressions of IL-6 and VEGF in the periodontal tissues of rats in the periodontitis group were significantly higher than those in the control group(all P<0.05),and the mean op-tical densities in the magnolol treatment group and topical+magnolol treatment group were significantly lower than those in the periodontitis group(all P<0.05),and the topical+magnolol treatment group was significantly lower than the topical treatment group(all P<0.05).Conclusion Magnolol can significantly reduce the level of inflammatory cytokines in perio-dontal tissues of rats with periodontitis,alleviate periodontal lesions,and improve the neoplastic state of alveolar bone.
