Intervention of decoction for warming the gallbladder on formation of endolymphatic hydrops via regulating p38 MAPK signaling pathway
Objective It is to investigate the effect of decoction for warming the gallbladder on formation of endolym-phatic hydrops via regulating p38 MAPK signaling pathway.Methods Thirty guinea pigs were randomly divided into control group,model group,low-dose,medium-dose and high-dose groups of decoction for warming the gallbladder,with 6 pigs in each group.The pigs of all groups except for the control group were treated with 6 μg/kg of desmopressin acetate by intrap-eritoneal injection for 10 days to establish models of endolymphatic hydrops.Starting from the 11th day,the ow-dose,me-dium-dose and high-dose groups of decoction for warming the gallbladder were gavaged with 6.1 g/kg,12.2 g/kg and 24.4 g/kg of decoction for warming the gallbladder,respectively,the control group and model group were gavaged with equal volume of double-distilled water.The guinea pigs were executed after 10 d of continuous gavage,and the positive expressions of in-flammatory factors[interleukin-1β(IL-1β),interleukin-6(IL-6)]and aquaporin 2(AQP2)in cochlear tissues were de-tected by immunohistochemistry,the expressions of AQP2 and p38 MAPK mRNA in cochlear tissues were detected by qRT-PCR,and the expressions of p38 MAPK,p-p38 MAPK proteins in cochlear tissues were detected by Western blot.Results Compared with the control group,the absorbance values of positive expressions of IL-1β,IL-6,AQP2 and relative expres-sions of AQP2 mRNA in cochlear tissues and p-p38 MAPK/p38 MAPK ratio of the model group were significantly increased(all P<0.05),and there was no significant change in the relative expression of p38MAPK mRNA(P>0.05);compared with the model group,the absorbance values of positive expressions of IL-1β,IL-6,AQP2 and relative expressions of AQP2 mRNA in cochlear tissues and p-p38 MAPK/p38 MAPK ratio of each dose group of decoction for warming the gall-bladder were significantly decreased(all P<0.05),and there was no significant change in the relative expression of p38MAPK mRNA(P>0.05).Conclusion Decoction for warming the gallbladder can effectively improve endolymphatic hydrops in guinea pigs,and its mechanism may be related to inhibiting P38 MAPK pathway and regulating the expression of AQP2.
