Protective effect of Jinlida granule combined with semaglutide on non-alcoholic fatty liver disease
Objective It is to explore the protective effect of Jinlida granule combined with semaglutide on obese mice with non-alcoholic fatty liver disease(NAFLD).Methods Thirty-two 6-week-old C57BL/6J male mice were selected,in which 8 mince were randomly selected as normal group fed with normal diet,and the other mice were fed with high-fat diet for 12 weeks to establish NAFLD models.The successfully modeled NAFLD mice were randomly divided into NAFLD group,semaglutide group,Jinlida combined with semiglutide group(combination group),with 8 mice in each group.The semaglutide group was given semiglutide 30 nmol/kg by intraperitoneal injection,the combination group was given Jinlida granules 1.5 g/kg by gavage on the basis of semaglutide by injection,the normal group and NAFLD group were given nor-mal saline by gavage,all once daily,continuously treated for 12 weeks.The body weight of mice in each group was recor-ded,and the levels of blood lipids[triacylglycerol(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C)],liver function indexes[alanine aminotransferase(ALT),aspar-tate aminotransferase(AST)]and fasting insulin(FINS)were detected.The histopathological changes of liver were ob-served by HE staining,oil red O staining,Masson staining and transmission electron microscopy.Results The body weight and serum levels of ALT,AST,TG,TC and LDL-C in the NAFLD group were significantly higher than those in the normal group(all P<0.05).Extensive diffuse hepatocyte steatosis,hepatocyte balloon-like degeneration,focal lymphocyte infil-tration,large amount of lipid deposition,mitochondrial swelling,mitochondrial ridge breakage and reduction,and mito-chondrial microautophagy were observed in the liver tissue.Compared with the NAFLD group,the body weight and serum levels of ALT,AST,TG,TC,LDL-C and FINS of mice in the semaglutide group and combination group were significantly decreased(all P<0.05),and the liver lesions were significantly alleviated.Compared with the semaglutide group,the de-creases of the above indexes were more significant in the combination group(all P<0.05),and the liver lesions were more milder.Conclusion Jinlida granules combined with semaglutide can significantly regulate lipid metabolism,protect liver function,alleviate insulin resistance,improve liver steatosis,lipid deposition,inflammatory cell infiltration and mitochon-drial microautophagy structure in NAFLD mice.
