Antibacterial effects of Chinese and Mongolian medicines on Helicobacter pylori in vitro
Objective It is to screen the single Chinese and Mongolian medicines with antibacterial effects on Helico-bacter pylori(H.pylori),to construct new antibacterial prescriptions,and to explore the therapeutic value of Chinese and Mongolian medicines on H.pylori.Methods The standard strain of H.pylori was cultured in vitro,and the H.pylori strains of the patients with current H.pylori infection were isolated and cultured in clinic.At the same time,biochemical reactions and H.pylori specific gene detection were used for identification.The sensitivity tests of standard and clinical H.pylori strains were carried out on Coptis chinensis,Scutellariae radix,Rheum palmatum,Clove,Houttuynia cordata,Astragalus membranaceus,Poria cocos,Panax notoginseng,ginger,Sophora flavescens,Chebulae Fruit and Dandelion were respec-tively carried out.The antibacterial diameter was measured by cross measurement method.The diameter of 15-20 mm was highly sensitive,and the diameter of 10-14 mm was moderately sensitive.According to the above experiments,the sensi-tive drugs were selected to construct a new prescription,and its antibacterial effect was further detected by drug sensitivity test,concentration gradient analysis and urease concentration measurement.Amoxicillin,clarithromycin,metronidazole,levofloxacin,rifampicin,tetracycline,gentamicin,and furazolidone were selected for drug sensitivity testing of H.pylori clinical strains to analyze the drug resistance and the susceptibility of H.pylori strains from drug-resistant patients to the new prescription.Results The standard strain of H.pylori is highly sensitive to Chebulae Fruit and Dandelion,moderately sensitive to Clove and Rheum palmatum.The clinical strain of H.pylori is highly sensitive to Dandelion,moderately sensi-tive to Houttuynia cordata,Panax notoginseng,Sophora flavescens,Scutellariae radix,Coptis chinensis,Astragalus mem-branaceus,Chebulae Fruit,Clove,Rheum palmatum and ginger.Dandelion,Chebulae Fruit,Clove,Rheum palmatum all had good antibacterial effects on standard strain and clinical strain of H.pylori,and the effect of Dandelion was the best.According to the results of sensitivity tests,a new prescription named Pinellia Yiwei granules was composed:Pinellia Tuber 15 g,Coptis chinensis 6 g,Scutellaria 10 g,Dry ginger 10 g,Salvia miltiorrhiza 10 g,Baked licorice 6 g,Jujube 15 g,Chebulae Fruit 10 g,Clove 6 g,Panax notoginseng powder 6 g,Dandelion 30 g,Lilium 30 g,Combined Spicebush Root 10 g,Houttuynia cordata 10 g,Cuttlefish 15 g.6 cases of H.pylori clinical strains were highly sensitive to Pinellia Yiwei granules,and 3 cases of H.pylori clinical strains and standard strains were moderately sensitive to Pinellia Yiwei granules.The minimum inhibitory concentration of Pinellia Yiwei granules was 0.5 g/mL,and the concentration of urease secreted by the standard and clinical strains of H.pylori were decreased significantly after the application of the granules used at this concentration compared with that before the application(all P<0.05).Antibiotic resistance of H.pylori clinical strains:7 cases of metronidazole,4 cases of levofloxacin,3 cases of clarithromycin,1 case of furazolidone,1 case of amoxicillin;Pinellia Yiwei granules was both highly sensitive to metronidazole and levofloxacin-resistant H.pylori clinical strains.Con-clusion Dandelion,Chebulae Fruit,Clove,Rheum palmatum have good inhibitory effects on both standard and clinical strains of H.pylori.The new prescription Pinellia Yiwei granules have significant inhibitory effect on standard and clinical strains of H.pylori,and also have inhibitory effects on some antibiotic resistant clinical strains of H.pylori.It is expected to become an adjuvant drug for eradicating H.pylori.
