清化饮对脾胃湿热型抑郁症大鼠色氨酸代谢通路及小肠病变的影响

目的 探讨清化饮对脾胃湿热型抑郁症大鼠色氨酸代谢通路及其小肠病变的影响。方法 取30 只SD雄性大鼠,随机选择5 只作为对照组,剩余大鼠以高脂高糖饮食和慢性温和性不可预知应激方法干预28d建立脾胃湿热型抑郁症模型。将造模成功大鼠随机分为模型组6 只、清化饮组6 只、舍曲林组6 只和清化饮+舍曲林组7 只。清化饮组给予清化饮27。5 g/kg灌胃,舍曲林组给予舍曲林5。21 mg/kg灌胃,清化饮+舍曲林组交错间隔1h灌胃清化饮和舍曲林,模型组和对照组给予等量生理盐水灌胃,均1 次/d,连续灌胃21 d。实验结束后,采用旷场实验评估大鼠行为学情况;ELISA法检测血清色氨酸、5-羟色胺(5-HT)、色胺水平,计算色胺/色氨酸、5-HT/色氨酸比值;HE染色法观察小肠组织病理形态。结果 实验结束后,模型组大鼠表现出现明显抑郁样表现,旷场实验中大鼠外围穿格数、直立次数均明显少于对照组(P均<0。05),各给药组大鼠抑郁样表现有所改善,其中清化饮组和清化饮+舍曲林组大鼠中心穿格数均明显多于模型组(P均<0。05),清化饮组外围穿格数明显多于清化饮+舍曲林组(P<0。05)。与对照组比较,模型组血清5-HT水平、色胺/色氨酸比值、5-HT/色氨酸比值均明显降低(P均<0。05),血清色氨酸水平明显升高(P<0。05),血清色胺水平无明显变化(P>0。05);小肠组织黏膜固有层间质淋巴细胞聚集,淋巴小结形成。与模型组比较,清化饮组和清化饮+舍曲林组血清5-HT水平、5-HT/色氨酸比值均明显升高(P均<0。05),清化饮组和清化饮+舍曲林组血清色胺水平、色胺/色氨酸比值及清化饮+舍曲林组血清色氨酸水平均明显降低(P均<0。05);舍曲林组血清5-HT水平、色胺/色氨酸比值、5-HT/色氨酸比值均明显升高(P均<0。05),血清色氨酸水平明显降低(P<0。05),血清色胺水平无明显变化(P>0。05);各给药组小肠组织炎症细胞浸润程度均明显减轻,其中清化饮+舍曲林组绒毛长度与隐窝深度之比变大。与舍曲林组比较,清化饮组、清化饮+舍曲林组血清色氨酸水平均明显升高(P均<0。05),血清色胺水平、色胺/色氨酸比值、5-HT/色氨酸比值均明显降低(P均<0。05)。结论 清化饮可能通过调控色氨酸代谢过程和提高血清 5-HT水平改善脾胃湿热型抑郁症大鼠抑郁症状,减轻小肠病理损伤,与舍曲林联合应用可以提高色氨酸水平,降低色胺水平及其与色氨酸的比值,从而增加色氨酸利用率,可以有效改善小肠消化和吸收功能。
Effects of Qinhuayin on the tryptophan metabolism pathway and small intestinal pathology in rats with depression of syndrome of spleen-stomach damp-heat
Objective It is to investigate the effects of Qinhuayin on the tryptophan metabolism pathway and small in-testinal pathology in rats with depression of syndrome of spleen-stomach damp-heat.Methods Thirty SD male rats were taken,in which 5 rats were randomly selected as the control group,and the remaining rats were treated with high-fat and high-sugar diet and chronic mild unpredictable stress for 28d to establish models of depression of syndrome of spleen-stom-ach damp-heat.The successfully modeled rats were randomly divided into 6 rats in the model group,6 rats in the Qinhuay-in group,6 rats in the sertraline group and 7 rats in the Qinhuayin+sertraline group.The Qinghuayin group was given Qin-ghuayin 27.5 g/kg by gavage,the sertraline group was given sertraline 5.21 mg/kg by gavage,and the Qinghuayin+ser-traline group was given Qinghuayin and sertraline at intervals of 1 h by gavage,while the model group and control group were given equal amounts of normal saline by gavage,all once daily,continuously treated for 21 days.At the end of the ex-periment,the behavioral conditions of the rats were assessed by open field test;the serum levels of tryptophan,5-hydroxytryptamine(5-HT),and tryptophan were detected by ELISA,and tryptophan/tryptophan and 5-HT/tryptophan ra-tios were calculated;the histopathological morphology of the small intestine were observed by HE staining.Results At the end of the experiment,the rats in the model group were obviously depressive,and their numbers of crossing periphery grid,standing upright were all significantly less than those in the control group(all P<0.05),and the depression of rats in the treatment groups was improved,in which the numbers of crossing center grid of rats in the Qinghuayin group and Qinghuay-in+sertraline group were significantly higher than that in the model group(all P<0.05),the number of crossing peripher-y grid of rats in the Qinghuayin group was significantly higher than that in the Qinghuayin+sertraline group(P<0.05).The serum level of 5-HT,tryptamine/tryptophan ratio,and5-HT/tryptophan ratio in the control group were all significantly lower(all P<0.05),while the serum level of tryptophan was significantly higher in the model group(P<0.05),there was no significant difference in the serum level of tryptamine between the two groups(P>0.05);interstitial lymphocytes in the mucosal lamina propria of small intestinal tissues were aggregated,and lymphoid nodules were formed in the model group.Compared with the model group,the serum level of 5-HT and 5-HT/tryptophan ratio were significantly higher in the Qinghuayin group and Qinghuayin+sertraline group(all P<0.05),while the serum level of tryptamine,tryptamine/trypto-phan ratio in the Qinghuayin group and Qinghuayin+sertraline group and serum level of tryptophan in the Qinghuayin+ser-traline group were significantly lower(all P<0.05);the serum level of 5-HT,tryptamine/tryptophan ratio,and 5-HT/tryptophan ratio in the sertraline group were all significantly higher(P<0.05),while the serum level of tryptophan level was significantly lower(P<0.05),and there was no significant difference in serum level of tryptamine between the two groups(P>0.05);the infiltration of inflammatory cell in small intestinal tissues of the treatment groups was significantly reduced,of which the ratio of the villi length to the crypts depth in the Qinghuayin+sertraline group became larger.Com-pared with the sertraline group,the serum level of tryptophan was significantly higher(P<0.05),and the serum level of tryptamine,tryptamine/tryptophan ratio,and 5-HT/tryptophan ratio were significantly lower in the Qinghuayin+sertraline group(all P<0.05).Conclusion Qinghuayin can improve the depressive symptoms and reduce the pathological damage of small intestine in rats with depression of syndrome of spleen-stomach damp-heat via regulating the metabolic process of tryp-tophan and increasing the serum level of 5-HT,and its combined application with sertraline can increase the serum level of tryptophan,decrease the level of tryptophan and its ratio to tryptophan,thus to increase the utilization rate of tryptophan,and effectively improve the digestive and absorptive functions of small intestine.
