Effect of electroacupuncture at Ningshen acupoint on NLRP3/Caspase-1/GSDMD pathway mediated-pyroptosis in rats with cerebral hemorrhage
Objective It is to investigate the neuroprotective effect of electroacupuncture at Ningshen acupoint on rats with cerebral hemorrhage via NLRP3/Caspase-1/gasdermin D ( GSDMD) pathway mediated pyroptosis.Methods Twenty-four male SD rats were randomly divided into normal group,sham operation group,cerebral hemorrhage group and cerebral hemorrhage+electroacupuncture group,with 6 rats in each group.The rats in the cerebral hemorrhage group and cerebral hemorrhage+electroacupuncture group were treated with double injection of intracerebral autologous arterial blood to pre-pare the models of cerebral hemorrhage.The cerebral hemorrhage+electroacupuncture group was treated with electroacu-puncture at the bilateral Ningshen acupoint for 30 min,once daily,continuously treated for 7 days.The sham operation group was only simulated the surgical trauma process without injecting autologous blood,the normal group was treated with no intervention.The degree of cerebral nerve function damage of rats in each group was evaluated by Longa score at 24 h af-ter surgery and;after intervention for 7 days,the rats in each group were executed,the water content of brain tissue was detected by wet and dry weight method;the pathologic morphology of cerebral tissue was observed by HE staining,the con-tents of interleukin-1β (IL-1β) and interleukin-18 (IL-18) in cerebral tissue were detected by ELISA,the protein expres-sions of and NLRP3,Caspase-1 p20,and GSDMD in cerebral tissue were detected by Western blot method,and the posi-tive protein expressions of NLRP3,Caspase-1,and GSDMD proteins in cerebral tissues were detected by immunohisto-chemistry.Results After 7 days of intervention,the neurobehavioral scores,contents of water,IL-1βand IL-18 in cerebral tissue,relative protein expressions of NLRP3,Caspase-1 and GSDMD,and the average integral optical density values of positive protein expressions of NLRP3,Caspase-1 and GSDMD in cerebral tissue of rats in the cerebral hemorrhage group were significantly higher than those in the normal group and sham operation group (all P<0.05),with edema and inflam-matory cell infiltration in the hemorrhage area and surrounding brain tissues;all of the above indexes were significantly low-er in the rats of the cerebral hemorrhage+electroacupuncture group than those in the cerebral hemorrhage group ( all P<0.05 ),and the edema and inflammatory cell infiltration in the hemorrhage area and surrounding brain tissues were signifi-cantly improved.Conclusion Electroacupuncture at the Ningshen acupoint can significantly relieve cerebral nerve and brain tissue injury in rats with cerebral hemorrhage,and the mechanism may be related to down-regulating the expression of pro-teins related to the NLRP3/Caspase-1/GSDMD signaling pathway to inhibit pyroptosis.
