摘要

目的:基于生物信息学分析糖尿病心肌病的潜在生物标志物.方法:糖尿病组及对照组小鼠心肌组织表达谱从基因表达数据库(gene expression omnibus,GEO)下载.应用基因本体(gene ontology,GO)和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)分析差异表达基因的功能和通路.通过基因/蛋白质相互作用检索工具(search tool for the retrieval of interacting genes/proteins,STRING)和Cytoscape软件用于构建蛋白质-蛋白质相互作用网络并筛选关键差异表达基因,通过人类蛋白质谱(human protein atlas,HPA)数据库筛选差异表达且在血液中可通过免疫法检测的细胞外蛋白基因.结果:共筛选出857个差异表达基因,主要富集的细胞过程为小鸟苷三磷酸(guanosine triphosphate,GTP)酶介导的信号调节、大鼠肉瘤蛋白(Rat Sarcoma,Ras)信号调节、蛋白质乙酰化.最终筛选出7个差异表达基因:CXC趋化因子配体10(CXC chemokine ligand 10,Cxcl10)、间质表皮转化因子(mesenchymal to epithelial transition factor,Met)、巨噬细胞集落刺激因子受体(feline mcDonough sarcoma receptor,Fms)样酪氨酸激酶 1(FMS-like tyrosine kinase 1,Flt1)、促红细胞生成素(erythropoietin,Epo)、白细胞介素 15(interleukin-15,Il15)、转铁蛋白受体(transferrin receptor,Tfrc)、β2 微球蛋白(beta-2 Microglobulin,B2m),编码可在血液中通过免疫法检测的细胞外蛋白.结论:Cxcl10、Met、Flt1、Epo、Il15、Tfrc、B2m或许可作为糖尿病心肌病筛查的潜在的生物标志物.

Abstract

Objective:To analyze the potential biomarkers of diabetic cardiomyopathy based on bioinformatics.Methods:The myocardial expression profiles of diabetic mice and control mice were downloaded from gene expression omnibus(GEO).Gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)were used to analyze the functions and pathways of differentially expressed genes.Search tool for the retrieval of interacting genes/proteins(STRING)and cytoscape were used to construct protein-protein interaction network and screen key differentially expressed genes.The differentially expressed genes encoding extracellular proteins that can be detected by blood-based immunoassay were screened by human protein atlas(HPA)protein annotation database.Results:A total of 857 differentially expressed genes were screened,and the enriched biological process were regulation of small GTPase mediated signal transduction,regulation of Ras protein signal transduction and protein acetylation.Seven genes encoding extracellular proteins,detectable by blood-based immunoassay,were selected:CXC chemokine ligand 10(Cxcl10),mesenchymal to epithelial transition factor(Met),FMS-like tyrosine kinase 1(Flt1),erythropoietin(Epo),interleukin-15(Il15),Transferrin receptor(Tfrc),and beta-2 microglobulin(B2m).Conclusions:Cxcl10,Met,Flt1,Epo,Il15,Tfrc,B2m might be potential biomarkers for screening diabetic cardiomyopathy.
