二次回归正交旋转组合设计优化伊宁红皮蒜鳞茎盘组织培养研究

[目的]研究植物激素对伊宁红皮蒜鳞茎盘不定芽生长的影响,为进一步完善大蒜的快繁体系提供依据.[方法]采用二次回归正交旋转组合实验,设计不同的IBA、NAA、6 - BA水平及其组合进行实验.[结果]影响伊宁红皮蒜鳞茎盘不定芽生长的三因素主次顺序是IBA> NAA>6- BA.其中IBA影响极显著,但其高浓度有负效应,在进一步实验时去掉了IBA,以降低对伊宁红皮蒜鳞茎盘不定芽生长的影响,6- BA单因素影响显著,为正效应.[结论]伊宁红皮蒜鳞茎盘不定芽生长最佳激素配比为:NAA浓度为1.799 mg/L,6 - BA浓度为7.50 mg/L,在此条件下每个伊宁红皮蒜鳞茎盘不定芽生长平均值可达22.92株.
Optimization of Tissue Culture in Xinjiang Red Garlic by Using Quadratic Regression Orthogonal Rotation Combination Design
[Objective] This study researched the effects of the plant hormones on Xinjiang red garlic bulb disc adventitious shoots growth in order to provide a theoretical basis for rapid propagation system of Xinjiang white garlic. [Method] The experiment adopted quadratic regression orthogonal rotation combination design and designed different IBA, NAA, 6 - BA level and their combination.[Result] Results showed that the effects on adventitious shoots growth were in the following order; IBA ? NAA ?6 - BA. The effect on IBA was very significant but its high concentration had negative effects, so in further experiments IBA was abolished in order to reduce interaction to Xinjiang red garlic bulb disc adventitious shoots growth effects. 6 -BA had obvious effects on the single - factor with positive effect. [ Conclusion ] The best hormone ration for Xinjiang red garlic bulb disc adventitious shoots growth was: NAA concentration was 4 mg/L, 6 - BA cone ulration was 7.5 mg/L. Under this condition Xinjiang red garlic disc adventitious shoots growth could get an average of 22.92.
