新疆核桃树腐烂病拮抗细菌的筛选及初步鉴定

[目的]核桃腐烂病已成为制约新疆核桃产业健康发展的重要病害之一,筛选拮抗菌株开展该病害的生物防治,有利于新疆生态健康果园的建设.[方法]采用平板分离、对峙培养、致畸菌丝形态观察、拮抗菌形态学、生理生化特性及分子生物学的鉴定方法.[结果]获得具有广谱拮抗效果的拮抗菌株XJAU-117,对核桃树腐烂病病原菌的拮抗效果达85%.通过形态学观察、生理生化特性和16s rDNA序列分析,鉴定为短小芽孢杆菌(Bacillus pumilus),拮抗菌株对核桃树腐烂病病原菌的抑制机理主要为使正常菌丝发生断裂、解体,无法正常生长.[结论]所获得的拮抗细菌对核桃树腐烂病病原菌表现出较好的拮抗作用,对生物防治核桃树腐烂病具有潜在的应用价值.
Isolation and Identification of the Antagonistic Bacteria against Walnut canker in Xinjiang
[Objective] Walnut canker disease (Cytospora chrysosperma) has become one of the important diseases which restricts the healthy development of walnut industry in Xinjiang.Biological control is beneficial to the construction of the ecological health of orchard in Xinjiang.[Method] Methods,such as plate screening,flat confrontation culture,antagonistic experiment and phylogenetic analysis of 16S rRNA were used.[Result] The mycelium shape and the bacterial colony,physiological and biochemistry experiments and determination of the sequence of 16S rDNA of the fungi had been done.The strain XJAU-117 was identified as Bacillus pumilus.[Conclusion] The result shows that the strain XJAU-117 with significant antagonistic effect to Cytospora chrysosperma could provide potential solution for biological control of Walnut canker.
