Analysis of Genetic Diversity and Construction of DNA Fingerprinting for Vitis Germplasm Based on SSR Markers
[Objective]The project aims to clarify the genetic diversity and establish the fingerprinting among the materials in this study, the present results provide valuable tools for the cultivar classification, germplasm identification and orientational breeding of dried grape.[Method]SSR markers were employed to clarify the genetic diversity and establish the fingerprinting of 44 grape (Vitis L.) genotypes suitable for processing of raisins.[Result]A total of 190 makers were scored from the 8 primers, which were screened out from 52 primers and demonstrated highly reproducible, clear and polymorphie band.190 DNA fragments were generated and all of them were polymorphic bands (the percentage of polymorphic band, PPB =100%).The ranges of PIC at 8 SSR loci of 44 cultivars were 0.686,8-0.964,0, with an average of 0.908 4.The clustering dendrogram was constructed by UPGMA method, the genetic similarity coefficient of the 44 accessions ranged from 0.63 to 0.92;all the materials were divided into 5 major groups while the similarity coefficient was 0.654, and to a certain extent reflect the evolutionary relationship among them.The primer pairs Vmc9a2.1, UDV-017, UDV-033 and UDV-041 were selected to establish the fingerprint of grape, which was able to distinguish all of the 44 accessions.[Conclusion]Using the method of SSR molecular markers could analyze the genetic relationship of the germplasms, and the four kinds of primers which were selected from the study could build its DNA fingerprint.
