Study on anti-PRRSV effect of 5-aminolevulinic acid
Porcine reproductive and respiratory syndrome(PRRS)is an acute,highly contagious disease caused by porcine reproductive and respiratory syndrome virus(PRRSV).5-aminolevulinic acid(5-ALA),as a natural amino acid,plays important antiviral function,and it inhibits replication of multiple viruses.However,whether 5-ALA can exert anti-PRRSV effect has not been reported,to our knowledge.In this study,the cytotoxicity of 5-ALA on PAM-KNU cells was detected by CCK-8 assay.The optimal method for and the optimal concentra-tion of 5-ALA to inhibit PRRSV propagation were determined by three modes of administration:prevention,direct killing and treatment.The results showed that the viral titers and cDNA copies were significantly down-regulated when the 5-ALA concentration was 1 mmol/L.Mean-while,the anti-PRRSV effect of prevention and direct killing of 5-ALA were more effective than the function of treatment.The effect of 5-ALA on PRRSV replication cycle was investigated by three periods:adsorption,internalization and early intracellular replication.The results showed that 5-ALA played a key role in the internalization and early intracellular replication period to inhibit PRRSV replication.Our study suggested that 5-ALA could inhibit PRRSV replication,which might serve as theoretical support for the prevention and treatment of PRRSV infection.
