Studies on the Methods of in vitro Evaluation of Black Rot-Resistance in Cabbage
In order to identify black rot resistance in cabbage rapidly,different inoculation methods,bacterium concentrations and seedling stages were compared with in vivo spray as the control.A good method was identified.Its procedure included the following steps: in vitro punching was made on the leaves of cabbage plants with 4-6 expanded leaves,and the leaves with a diameter of 1.5 cm were inoculated with a bacterium suspension with a concentration of 1.0×10^8 cfu/mL.Then the leaves were transferred to Petri plates containing wet filter paper and cultured at 28oC in an incubator.The colonies on the inoculated leaves were observed 5 days after inoculation.The identification result was consistent with that of in vivo spray.It is concluded therefore that this method can be used in resistance source selection,variety/line resistance identification and screening for resistant mutants.
