Effect and mechanism of Dendrobium officinale on atherosclerosis in ApoE-/-mice
Objective To investigate the effect and mechanism of Dendrobium officinale on atherosclerosis in ApoE-/-mice.Methods Atherosclerosis was induced in ApoE-/-mice model by high-fat diet.The mice were then randomly assigned to receive either saline(10 mL/kg),Shexiang Baoxin Pill solution(20 mg/kg),low-dose Dendrobium officinale solution(10 g/kg),or high-dose Dendrobium officinale solution(30 g/kg),this formed the model group,positive drug group,low-dose group,and high-dose group,with 8 mice in each group.Eight ApoE-/-mice fed with a normal diet were given saline(10 mL/kg)as the control group.After 4 weeks of treatment,histopathological changes in the thoracic aorta tissue of the mice were observed using hematoxylin-eosin(HE)staining.Levels of triglycerides(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)in the mice were measured.Quantitative fluorescence PCR was used to detect the expression of ATP-binding cassette transporter A1(ABCA1),ATP-binding cassette transporter G1(ABCG1)and peroxisome proliferator-activated receptor gamma(PPARγ).Results Compared with the control group,lipid plaques were formed in the inner artery of the model group,with rhomboid cholesterol crystals,inflammatory cell infiltration and a large number of foam cells gathering.Compared with the model group,the positive drug group,low-dose group,and high-dose group showed reduced foam cell deposition and alleviated atherosclerotic lesions in the thoracic aorta.Compared with the control group,TG,TC,LDL-C levels in the model group were higher,HDL-C level was lower,ABCA1,ABCG1,PPARγ expression levels were lower,and the differences were statistically significant(P<0.05).Compared with the model group,TG and TC levels decreased,and HDL-C levels increased in the positive drug group,low-dose group,and high-dose group(P<0.05).The expression levels of ABCA1,ABCG1,and PPARγ were higher in the positive drug group and high-dose group compared to the model group,with statistically significant differences(P<0.05).Conclusion Dendrobium officinale can improve blood lipid levels and alleviate atherosclerotic lesions in ApoE-/-mice,potentially by influencing the expression of ABCG1,ABCA1 and PPARγ.
