目的 探究鸦胆子结合吡柔比星对膀胱癌细胞的作用.方法 选取2023年4-5月期间黑龙江省医院泌尿科1株人BIU-87膀胱癌细胞株为研究对象,细胞培养后,将其接种于96孔板中,以随机数表法分为5组,每组细胞均进行3个平行样本,每个样本细胞均设置5个复孔(每组共计15个样本量),将5组细胞以不同浓度鸦胆子油乳(0、5、10、20、30 mL/L)与吡柔比星(5 µmol/L)联合作用,分析不同浓度鸦胆子油乳对膀胱癌细胞增殖抑制作用、细胞凋亡及细胞周期影响,并检测联合用药对核因子κB(Nuclear Factor κB,NF-κB)影响、对细胞中三磷酸腺苷(Adenosine Triphosphate,ATP)结合转运蛋白G家族成员 2(ATP-binding Cassette TransporerG2,ABCG2)表达情况影响.结果 鸦胆子油乳浓度升高,BIU-87细胞增殖抑制率、坏死率、凋亡率逐渐升高,差异有统计学意义(P均<0.05);鸦胆子油乳浓度为5 mL/L时细胞增殖抑制率、细胞坏死率显著提升,浓度≥10 mL后,细胞凋亡率显著提升,差异有统计学意义(P均<0.05);随鸦胆子油乳浓度升高,BIU-87细胞中S期细胞占比降低,C0/C1期细胞占比升高,差异有统计学意义(P均<0.05);随鸦胆子油乳浓度升高,细胞质中NF-κB表达量逐渐下降,细胞核中NF-κB表达量逐渐升高,细胞膜表面ABCG2表达量下降,差异有统计学意义(P均<0.05).结论 鸦胆子油乳结合吡柔比星,可抑制BIU-87膀胱细胞株增殖,低浓度可促进细胞坏死,高浓度可促进细胞凋亡,并促进NF-κB从细胞质转入细胞核中,抑制ABCG2表达,其效果均存在剂量依赖性.
Effect of Brucea Javanica Combined with Pirarubicin on Bladder Cancer Cells
Objective To explore the effect of brucea javanica combined with pirarubicin on bladder cancer cells.Methods A human BIU-87 bladder cancer cell line from Department of Urology of Heilongjiang Provincial Hospital during April to May 2023 was selected as the research object.After cell culture,the cell lines were inoculated into 96-well plates and divided into 5 groups by random number table method.Three parallel samples were taken from each group of cells,and five multiple pores were set in each sample(15 samples in total for each group).Five groups of cells were treated with different concentrations of brucea javanica oil emulsion(0,5,10,20,30 mL/L)combined with pi-rarubicin(5 µmol/L).The effects of different concentrations of brucea javanica oil emulsion on proliferation inhibition,apoptosis and cell cycle of bladder cancer cells were analyzed.The effects of combined administration on nuclear fac-tor κB(NF-κB)and the expression of adenosine triphosphate(ATP)combined with ATP-binding cassette transpor-erG2(ABCG2)in cells were detected.Results The proliferation inhibition rate,necrosis rate and apoptosis rate of BIU-87 cells were increased with the increase of brucea javanica oil emulsion concentration,the difference was statisti-cally significant(all P<0.05).The cell proliferation inhibition rate and cell necrosis rate were significantly increased when the concentration of brucea javanica oil was 5 mL/L,and the apoptosis rate was significantly increased when the concentration was greater than 10 mL,the difference was statistically significant(all P<0.05).The proportion of S phase cells in BIU-87 cells decreased and the proportion of C0/C1 phase cells increased with the increase of the con-centration of brucea javanica oil emulsion,the difference was statistically significant(all P<0.05).As the concentration of brucea javanica oil emulsion increases,in the cytoplasm the expression level of NF-κB gradually decreases,in the nucleus the expression level of NF-κB gradually increased,while the expression level of ABCG2 on the cell mem-brane surface decreased,the difference was statistically significant(all P<0.05).Conclusion Brucea javanica oil emul-sion combined with pirarubicin can inhibit the proliferation of BIU-87 bladder cell line,low concentration can pro-mote cell necrosis,high concentration can promote cell apoptosis,and promote the transfer of NF-κB from cytoplasm to nucleus,and inhibit the expression of ABCG2 in a dose-dependent manner.
