A Study on the Application of Semi-nested Fully Automated Real-time Fluorescent Quantitative PCR Assay and Smear Antacid Staining Method in Early Detection of Active Pulmonary Tuberculosis
Objective To evaluate the efficacy of the application of semi-nested fully automated real-time fluorescent quantitative PCR assay with smear antacid staining method in the early detection of active pulmonary tuberculosis.Methods 90 patients with suspected pulmonary tuberculosis attending the Pingyi County Tuberculosis Control Center between September 2022 and September 2023 were included as the study objects.The sputum specimens of 90 sus-pected TB patients were tested simultaneously by Xpert MTB/RIF technique,smear antacid staining test and simple method solid culture method,and the test results were compared.Results Using the solid culture results as the gold standard,the sputum culture positive rate was 85.56%and the negative rate was 14.44%.The Xpert MTB/RIF posi-tive rate was 84.44%and the negative rate was 15.56%.Smear antacid staining test positive rate 48.88%,negative rate 51.11%.The Xpert MTB/RIF test had a higher sensitivity of 97.40%,accuracy of 96.67%,positive predictive value of 98.68%,and negative predictive value of 85.71%than the smear antacid staining method test of 50.64%,52.22%,88.64%,and 17.39%,the differences were statistically significant(x2=43.768,46.722,5.922,22.546,all P<0.05).Conclusion Compared with the smear antacid staining method,the application of semi-nested fully automated real-time fluorescence quantitative PCR assay(Xpert MTB/RIF)in the early detection of active tuberculosis is more efficacious.
