Lycorine Inhibits the Invasion and Migration of Cervical Cancer Caski Cells By Regulating Wnt/β-catenin Signaling Pathway
Objective To investigate the effect of lycorine on invasion and migration of Caski cells of cervical cancer and its possible mechanism.Methods Cervical cancer Caski cells cultured in the laboratory of School of Basic Medi-cine of Jimusi University from June to December 2023 were selected and inoculated into 96-well petri dishes.After being attached to the wall for 2 h,the serum-free 1640 culture medium was replaced.The untreated Caski cells were used as the control group.Caski cells added with 2,4 and 8 ug/mL lycorine were divided into low concentration ex-perimental group,medium concentration experimental group and high concentration experimental group,and 5 com-pound pores were set in each group.After incubation for 12,24 and 48 h,the inhibitory effect of lycorine on prolifera-tion of Caski cells and its effect on invasion and migration of Caski cells were detected.The mRNA expression levels of c-Myc and G1/S-specific Cyclin D1(Cyclin D1)were detected by RT-PCR.The expression levels of β-catenin,c-Myc and Cyclin D1 were detected by western blot.Results Compared with the control group,lycorine significantly inhibited the proliferation of Caski cells in a time-concentration dependent manner.The invasion and migration num-bers of Caski cells in the control group were 610.20±36.63 and 220.00±22.26,which were higher than 367.60±28.14 and 60.40±9.65 in the experimental group,and the differences were statistically significant(t=11.744,14.709,both P<0.05).Compared with control group,mRNA and protein expressions of β-catenin,c-Myc and Cyclin D1 in experimen-tal group were lower,and the differences were statistically significant(all P<0.05).Conclusion Lycorine may inhibit the proliferation of Caski cells by targeting Wnt/β-catenin signaling pathway.
