为了建立乌奴龙胆的组培快繁体系,该研究选用采自西藏自治区拉萨市堆龙德庆区羊达乡境内的乌奴龙胆植株进行组培快繁实验.实验对乌奴龙胆的合适外植体、HgCl2溶液最佳消毒时间、最佳不定芽诱导分化培养基、最佳生根培养基进行了筛选.结果表明:带节茎段适合作为乌奴龙胆组培实验中的外植体,叶片块不适合作为外植体;HgCl2溶液最佳消毒时间为6min,平均未污染率为66.67%±3.33%,平均成活率为63.33%±3.33%;最佳不定芽诱导分化培养基为:P7处理组(MS+0.1g/L肌醇+30g/L蔗糖+5g/L琼脂粉+2.0mg/L 6-BA+1.0mg/L IAA pH5.6),平均芽增殖倍数为2.47±0.12,平均芽长度为17.07±1.56mm,平均未玻璃化率为80.00±5.57%;最佳生根培养基为:T5 处理组(1/2MS+0.1g/L肌醇+30g/L蔗糖+5g/L琼脂粉+0.3mg/L NAA+0.3mg/L IBA pH5.6),平均生根率为100%,平均生根数为3.63±0.09条,平均根长度为39.03±0.30mm,平均根健康率为85.27%±1.22%.
In order to establish the rapid propagation system of Gentiana urnula,the research selected Gentiana urnula plants collected from Yangda Village,Duilong Deqing District,Lhasa,Xizang Autonomous Region for the rapid propa-gation experiment of tissue culture.The most suitable explants,the optimal disinfection time of HgCl2 solution,the opti-mal adventitious bud inducd differentiation medium,and the best rooting culture medium for Gentiana urnula were screened.The results showed that the stem segment with nodes was suitable as an explant in the tissue culture experi-ment of Gentiana urnula,while the leaf block was not suitable as explants.The optimal disinfection time for HgCl2 so-lution is 6 minutes,with an average uncontaminated rate of 66.67%±3.33%and an average survival rate of 63.33%± 3.33%.The optimal adventitious bud induced differentiation medium was as follows of P7 treatment group(MS+0.1g/L inositol+30g/L sucrose+5g/L agar powder+2.0mg/L 6-BA+1.0mg/L IAA pH 5.6),with an average bud proliferation multiple of 2.47±0.12,an average bud length of 17.07±1.56mm,and an average unvitrified rate of 80.00±5.57%.The optimal rooting culture medium was as follows ofT5 treatment group(1/2MS+0.1g/L inositol+30g/L sucrose+5g/L agar powder+0.3mg/L NAA+0.3mg/L IBA pH 5.6),with an average rooting rate of 100%,an average rooting number of 3.63±0.09,an average rooting length of 39.03±0.30mm,and an average rooting health rate of 85.27%±1.22%.
