墨脱野生金线莲组培快繁体系研究

Study on the rapid propagation system of tissue culture of Motuo wild Anoectochilus roxburghii

为了建立墨脱野生金线莲的组培快繁体系,该研究选用采自西藏自治区林芝地区墨脱县格林村的野生金线莲植株进行组培快繁实验.结果表明,在选用的3种材料中,叶片块不适合作为外植体,带节茎段和根适合作为外植体,HgCl2溶液最佳消毒时间为10min.愈伤组织和不定芽最佳诱导培养基为A5处理组(MS+0.1g/L肌醇+30g/L蔗糖+5g/L琼脂粉+1mg/L IBA+0.5mg/L NAA+2mg/L 6-BA),出愈率为73.33%,芽增殖倍数为8.87,平均芽长为13.30mm.无根苗最佳诱导生根培养基为B7处理组(1/2MS+0.1g/L肌醇+30g/L蔗糖+5g/L琼脂粉+0.5mg/L NAA+1mg/L IBA),平均根数为4.20,平均根长为26.43mm.
In order to establish a rapid propagation system for the tissue culture of Motuo wild Anoectochilus rox-burghii,the research selected wild Anoectochilus roxburghii plants collected from Gelin Village,Motuo County,Ny-ingchi Prefecture,Xizang Autonomous Region to conduct the corresponding experiment.The results showed that among the three selected materials,leaf blocks were not suitalbe as explants,and the stem segments and the roots with nodes were suitable as explants,while the optimal disinfection time of HgCl2 solution was 10 minutes.The opti-mal induction medium for callus and adventitious buds is A5 treatment group(MS+0.1g/L inositol+30g/L sucrose+5g/L agar powder+1mg/L IBA+0.5mg/L NAA+2mg/L 6-BA),the healing rate was 73.33%,the bud proliferation factor was 8.87,and the average bud length was 13.30mm.The optimal rooting medium for rootless seedlings was B7 treat-ment group(1/2MS+0.1g/L inositol+30g/L sucrose+5g/L agar powder+0.5mg/L NAA+1mg/L IBA),with an average root number of 4.20 and an average root length of 26.43mm.
