温针联合神经电刺激对三叉神经痛大鼠三叉神经节P2Y2受体及电压门控性钾通道表达的影响

目的:观察温针联合神经电刺激对三叉神经痛大鼠三叉神经节P2Y2受体及电压门控性钾通道表达的影响。方法:选取30只健康成年雄性SD大鼠,按随机数字表法分为对照组、模型组和实验组各10只。模型组和实验组采用颧骨下入路眶下神经缩窄术制备三叉神经痛大鼠模型。对照组只暴露框下神经而不做结扎。对照组和模型组不做任何治疗,实验组采用温针联合神经电刺激治疗。测定各组大鼠的痛阈值;采用HE染色检测三叉神经节组织学结构,采用免疫组化方法检测降钙基因相关肽(CGRP)浓度变化;采用RT-PCR法及Western Blot法检测3组大鼠三叉神经节内P2Y2受体及电压门控性钾通道表达。结果:与对照组比较,模型组大鼠术后14、28 d痛阈值显著降低(P<0。05);与模型组比较,实验组大鼠术后14 d和28 d痛阈值升高(P<0。05)。与对照组比较,模型组大鼠三叉神经节CGRP蛋白、P2 Y2受体表达和血清IL-1β、IL-6、TNF-α水平均升高(P<0。05);与模型组比较,实验组大鼠三叉神经节CGRP蛋白、P2 Y2受体表达和血清IL-1β、IL-6、TNF-α水平均降低(P<0。05)。与对照组比较,模型组大鼠三叉神经节中Kv1。4、Kv3。4、Kv4。2、Kv4。3 mRNA水平均降低(P<0。05);与模型组比较,实验组三叉神经节中Kv1。4、Kv3。4、Kv4。2、Kv4。3 mRNA水平升高(P<0。05)。结论:温针联合神经电刺激能够通过抑制CGRP的表达及P2Y2受体,促进电压门控性钾通道相关蛋白表达,并降低血清炎性因子,从而缓解三叉神经痛大鼠的疼痛,为三叉神经痛的治疗提供新思路。
Effect of Warm Needle Combined with Electrical Nerve Stimulation on Expression of P2 Y2 Receptors and Voltage-Gated Potassium Channels in the Trigeminal Ganglion of Rats with Trigeminal Neuralgia
Objective:To observe the effect of warm needle combined with electrical nerve stimulation on expression of P2 Y2 receptors and voltage-gated potassium channels in the trigeminal ganglion of rats with trigeminal neuralgia.Methods:A total of 30 healthy adult male SD rats were selected and divided into the control group,the model group,and the experiment group according to the random number table method,with 10 rats in each group.The model group and the experiment group used the infraorbital nerve constriction surgery via the zygomatic bone approach to prepare a rat model of trigeminal neuralgia.The control group only exposed the infraclavicular nerve without ligation.The control group and the model group did not receive any treatment,and the experiment group was treated with a combination of warm needle and electrical nerve stimulation.Measured the pain threshold of each group of rats;HE staining was used to observe the histological structure of the trigeminal ganglion,and immunohistochemistry was used to detect changes in the concentration of calcitonin gene-related peptide(CGRP);RT-PCR and Western Blot were used to detect the expression of P2 Y2 receptors and voltage-gated potassium channels in the trigeminal ganglia of rats in the three groups.Results:Compared with those in the control group,the pain threshold of rats in the model group was significantly reduced at 14 and 28 days after surgery(P<0.05);compared with those in the model group,the pain threshold of rats in the experiment group was increased at 14 and 28 days after surgery(P<0.05).Compared with those in the control group,the expression of CGRP protein and P2 Y2 receptor in the trigeminal ganglion,as well as the levels of serum IL-1 β,IL-6,and TNF-α of rats in the model group were all increased(P<0.05);compared with those in the model group,the experiment group showed a decrease in the expression of CGRP protein and P2 Y2 receptor in the trigeminal ganglion,as well as levels of IL-1 β,IL-6,and TNF-α in serum(P<0.05).Compared with those in the control group,the mRNA levels of Kv1.4,Kv3.4,Kv4.2,and Kv4.3 in the trigeminal ganglia of rats in the model group were all reduced(P<0.05);compared with those in the model group,the mRNA levels of Kv1.4,Kv3.4,Kv4.2,and Kv4.3 in the trigeminal ganglia in the experiment group were increased(P<0.05).Conclusion:Warm needle combined with electrical nerve stimulation can alleviate the pain of trigeminal neuralgia rats by inhibiting the expression of CGRP and P2 Y2 receptors,promoting the expression of voltage-gated potassium channel related proteins,and reducing serum inflammatory factors,providing new ideas for the treatment of trigeminal neuralgia.
