间断性人工重力可减轻模拟失重大鼠股动脉血管平滑肌细胞的凋亡

摘要
目的 探讨间断性人工重力(intermittent artificial gravity,IAG)对模拟失重大鼠股动脉血管平滑肌细胞(femoral artery smooth muscle cells,FASMC)凋亡及血管重塑的作用.方法 将 30只 8周龄的SD大鼠随机分为3组,每组 10只,包括对照组(control group,CON)、模拟失重组(tail-suspended group,SUS)及模拟失重大鼠每日站立 1h的间断性人工重力组(standing group,STD).用M30及TUNNEL染色法检测大鼠FASMC的凋亡情况;用Western blot法检测各组大鼠股动脉组织中蛋白Caspase-3、Bad、FasL、Bcl2及增殖相关蛋白PCNA的表达.结果 与CON组比较,SUS组和STD组大鼠FASMC M30及TUNNEL染色阳性率显著增加(P<0.05);STD组M30及TUNNEL染色阳性率较SUS组显著减少(P<0.05).SUS组大鼠股动脉组织蛋白Bad、FasL及Caspase-3的表达较CON组和STD组显著增多(P<0.05),STD组Bad、FasL及Caspase-3的表达较CON组显著增加(P<0.05).STD组凋亡抑制因子Bcl2及增殖相关蛋白PCNA的表达较CON组及SUS组皆显著增加(P<0.05).结论 IAG对抗可有效减轻模拟失重致FASMC的凋亡,表明IAG对抗在失重引起的大鼠股动脉血管适应性重建中发挥重要作用.
Abstract
AIM To explore the effects of intermittent artificial gravity(IAG)on apoptosis of femoral artery vascular smooth muscle cells(FASMCs)in rats from 3-week simulated microgravity.METHODS Thirty Sprague Dawley rats were randomly separated into three groups with ten rats in each group:control group(CON),tail-suspended group(SUS)and standing group(STD).SUS rats were used to simulate the effects of microgravity and homochronous countermeasure(STD,daily 1h of-Gx gravitation)was used to simulate the effects of intermittent artificial gravity(IAG).Apoptosis of FASMCs was assessed by M30 and TUNNEL staining,and the expressions of Bad,FasL,Caspase-3,Bcl2 and PCNA in femoral artery tissues were detected by Western blot.RESULTS Compared with those in CON group,M30 and TUNNEL-positive FASMCs in SUS group and STD group increased significantly(P<0.05)and those in STD group decreased significantly(P<0.05)compared with those in SUS group.Compared with those in CON group and STD group,the expressions of Bad,Fasl and Caspase-3 in SUS group up-regulated significantly(P<0.05)and those in STD group down-regulated obviously compared with those in SUS group.While the expressions of Bcl2 and PCNA in STD group enhanced significantly,compared with those in CON group and SUS group,the expression of either Bcl2 or PCNA in SUS group only had an up-regulating tendency,respectively.CONCLUSION IAG effectively alleviates the increase of apoptosis of FASMCs from simulated rats.These findings further indicate that IAG may play an important role in the adaptive remodeling of vessels induced by microgravity.
