Isolation and screening of anti-pain scorpion peptides
The Buthusmartensii Karsch is one of the important Chinese medicinal materials in China.Its venom is rich in modulating molecules of various ion channels and receptors,which has therapeutic effects on pain,epilepsy and other diseases.Therefore,the isolation,screening and identification of complex peptides in scorpion toxin have been the focus of research and development of scorpion toxins.In order to isolate and screen the scorpion venom polypeptide with analgesic effects,four scorpion venom peptide components were separated and purified by reversed-phase high-performance liquid chromatography,named BmKⅡ-10,BmKⅡ-11,BmKⅡ-16 and BmKⅡ-24,and their molecular weights were 7 530.8 Da,7 366.0 Da,5 949.6 Da and 6 852.4 Da,respectively.A pain model was constructed to examin the effects of scorpion venom polypeptide components on pain in mice.The results showed that BmKⅡ-10 and BmKⅡ-11 may be the pain-causing components in scorpion venom,while BmKⅡ-16 had a significant analgesic effect and could effectively inhibit spontaneous pain and mechanical pain caused by formalin and carrageenan.The peptide separation and purification method used in this study can effectively obtain a variety of peptide components in scorpion toxin,and further verify that some components are closely related to pain,which provides theoretical and experimental basis for the development and application of pain drugs.
