Preliminary Genotoxic Study of Trollius Chinensis Bunge
Objective To evaluate the genetic toxicity of Trollius Chinensis Bunge,so as to provide a toxicological basis for its use in clinical treatment and drug development.Methods Lyophilised Salmonella Typhimurium histidine-defi-cient strains TA97a,TA98,TA100,TA102 and TA1535 were selected,and experimental group,verification group,un-treated control group,purified water control group,DMSO control group and positive control group were set up for each strain.A bacterial revert mutation test was used,where 0.1 mL of Trollius Chinensis Bunge solution was added to each dish of the test group at a mass concentration equivalent to 5 000,1 563,488,153,and 48 μg/dish,respectively.The stand-ard mutagen used in the positive control group were selected according to the sensitivity of the strains,and were Dexon(50μg/dish),2-Aminofluorene(2-AF,10 μg/dish),Methyl methanesulfonate(MMC,0.5 μL/dish),1,8-Dihydroxyanthraqui-none(Dionone,50 μg/dish),Sodium azide(NaN3,0.25 μg/dish),and 2-Aminoanthracene(2-AA,1 μg/dish),each of which was prepared with dimethylsulfoxide as a solvent.Amino anthracene(2-AA,1 μg/dish),and each positive was for-mulated with dimethyl sulfoxide as solvent.In the verification test,the dosages were 8,40,200,1 000 and 5 000 μg/dish.Using the liver microsomal enzyme S9 as a metabolic activation system,the number of revertant colonies of 5 strains of Salmonella typhimurium were counted.Results Compared with the untreated control group,the tested group,puri-fied water control group and DMSO control group,the number of revertant colonies did not increase by more than 1 times,and there was no dose-response relationship,and no repeatable positive result at any test point.The number of revertant colonies induced by each standard mutagen in the positive control group was more than twice that in the untreated control group.With or without metabolic activation system,the strain TA97a,TA98,TA100,TA102,TA1535 did not induce gene mutation,and no genotoxicity was observed.Conclusion Under the experimental conditions,Trollius Chinensis Bunge has no mutagenicity in this test.
