Analysis of short term efficacy of alcohol extract of Melastoma dodecandrum Lour against duck hepatitis B virus infection
Objective To investigate the in vivo anti-duck hepatitis B virus(DHBV)and hepatoprotective effects of the alcohol extract of Melastoma dodecandrum Lour.Methods Animal models of DHBV infection were established.And they were randomly divided into 6 groups(with 10 ducks in each group):high,medium and low dose groups of alcohol extract of Melastoma dodecandrum Lour,placebo group,lamivudine group,and non-infected group,and intragastric administration was performed once a day for 28 days.Venous blood were collected from each group of ducks before administration(T0),7 days after administration(T7),14 days(T14),21 days(T21),28 days(T28)and 5 days after drug withdrawal(P5).And then,venous blood of each group of ducks was collected to detect duck hepatitis B virus(DHBV DNA)levels in duck serum samples by real-time fluorescent quantitative PCR,and duck hepatitis B surface antigen(DHBsAg)and duck hepatitis B e antigen(DHBeAg)by enzyme-linked immunosorbent assay(ELISA).ALT and AST levels were detected,and the ducks were killed and their livers were dissected and removed 6 days after drug withdrawal(P6),and pathological sections of their livers were observed.Results At T0,there was no statistically significant difference in baseline indicators(P>0.05).No decrease in DHBV DNA load or DHBsAg OD value was observed in all dose groups of alcohol extract of Melastoma dodecan-drum Lour(P>0.05),and its effect on AST was not significant.The ALT and OD value of DHBeAg in each dose group of the alcohol extract of Melastoma dodecandrum Lour decreased(P<0.05).The liver pathological results showed that the de-gree of lesion in the high-dose group of the alcohol extract of Melastoma dodecandrum Lour was lower than that in other dose groups(P<0.05).Conclusion No inhibitory effects on DHBV and DHBsAg are found in the alcohol extract of Melastoma dodecandrum Lour,and its effect on AST is not significant.The alcohol extract of Melastoma dodecandrum Lour can inhibit DHBeAg and reduce ALT levels in all dose groups.Those in the high-dose group can improve liver pathological damage and has a certain protective effect on the liver.
