Study of Sema4D/interleukin-6 axis mediated interaction between neutrophils and endothelial cells in Kawasaki disease
Objective To explore the role of semaphorin 4D(Sema4D)/interleukin-6(IL-6)axis in interaction between neu-trophils and human coronary artery endothelial cells(HCAECs)in Kawasaki disease(KD).Methods A total of 55 patients with acute KD were selected as KD group,and 30 healthy children were selected as HC group.The concentrations of Sema4D and IL-6 in peripheral blood of the two groups were detected by ELISA,and the expressions of Sema4D on the surface of neutrophils were detected by flow cytometry;Sema4D was used to stimulate endothelial cells in vitro and the expressions of IL-6 were detected;after treating neutrophils with endothelial cell-conditioned medium,the shedding of Sema4D was detected.Results Compared with healthy children,Sema4D and IL-6 in peripheral blood in the acute phase of KD significantly increased,while the Sema4Dhigh neu-trophils significantly decreased,and difference was statistically significant(P<0.05).Compared with unstimulated group,the ex-pressions of IL-6 in endothelial cells stimulated by Sema4D significantly increased,and difference was statistically significant(P<0.05).Compared with unstimulated group,Sema4D-treated group's endothelial cell-conditioned medium significantly increased the shedding of Sema4D in neutrophils(P<0.01).Conclusion There may be a Sema4D/IL-6 axis mediated interaction between neu-trophils and endothelial cells during the process of coronary artery disease in KD.
