模型化研究Cyclin D1介导的胰岛素对糖代谢的调节

胰岛素(Insulin)通过细胞周期蛋白D1(Cyclin D1)调节糖代谢的物理机制,一直缺乏系统完整的理解.基于Hill动力学与Michaelis-Menten方程,建立理论模型研究Cyclin D1介导的Insulin对糖代谢的调节作用,研究发现,Insulin通过刺激AKT抑制GSK3,促进Cyclin D1表达提升.形成的Cyclin D1-CDK4复合物,改变组蛋白乙酰转移酶(GCN 5)的活性及磷酸化水平,实现对糖代谢重要参与酶PCK1的抑制调节,进而调控糖转化代谢.理论结果符合实验,进一步系统完整地揭示了Cyclin D1介导的Insulin对糖代谢调节的调控机理,确认了Cyclin D1对糖代谢的调控作用,可为设计阻断Cyclin D1异常导致的病变治疗方案提供理论依据.
Modelling Study on the Regulation of Insulin on Glucose Metabolism Mediated by Cyclin D1
Insulin regulates glucose metabolism through Cyclin D1,which has been lack of systematic and complete understanding.Based on Hill dynamics and Michaelis-Menten equation.In this paper,we built a theoretical model to study the regulatory effect of Insulin mediated by Cyclin D1 on glucose metabolism.We found that Insulin inhibited GSK3 by stimulating AKT and promoted the expression of Cyclin D1.The formed Cyclin D1-CDK4 complex can change the activity and phosphorylation level of GCN5,real-ize the inhibition and regulation of the enzyme PCK1,which is an important participant in glucose metabolism,and then regulate glu-cose metabolism.The theoretical results are consistent with the experiment.It further systematically and completely revealing the regu-latory mechanism of insulin on glucose metabolism mediated by Cyclin D1,confirming the regulatory effect of Cyclin D1 on glu-cose metabolism,and providing theoretical basis for the design of the treatment plan for blocking the pathological changes caused by the abnormality of Cyclin D1.
