The Application of Automated Magnetic Bead Pretreatment in the Detection of Catecholamines and Their Metabolites in Plasma by Liquid Chromatography Tandem Mass Spectrometry
Objective To investigate the effectiveness of automatic magnetic bead pretreatment in the detecting catecholamines and their metabolites in plasma by liquid chromatography tandem mass spectrometry(LC-MS/MS).Methods With the use of Automated magnetic bead method to extract catecholamines[norepinephrine(NE),epinephrine(E),dopamine(DA)]and their metabolites[methoxynorepinephrine(NMN),methoxyepinephrine(MN),3-methoxytyramine(3-MT)]from plasma samples and the use of LC-MS/MS to detect,the linearity,accuracy,precision,and matrix effects(ME)of this detection method were analyzed.Results The linear correlation coefficients r2 of NE,E,DA,NMN,MN and 3-MT detection were greater than 0.99,and the deviation range of each concentration value from the standard value was 0%~14.84%,all of which were less than 15%.The intra-batch coefficient of variation(CV)of low,medium and high values of NE,E,DA,NMN,MN and 3-MT was 0%~13.480%,and the inter-batch CV was 1.260%~13.590%,which met the requirements of both intra-batch and inter-batch CV≤15%.The ME of NE,E,DA,NMN,MN,and 3-MT were all in the range of-20%to 20%,indicating that the effect of ME was small.Conclusion With the use of LC-MS/MS for detecting catecholamines and their metabolites,and the use of automated magnetic bead method for plasma sample pretreatment,the linearity,accuracy,precision,and ME detection results for each indicator met industry standards.
