Clone and Quantitative Analysis by Real-Time PCR of Plant Architecture Traits Gene ZmDwarf4 in Maize
The Dwarf4 gene in maize which was homologous to the Dwarf4 gene in rice, and that was successfully cloned from inbred line Shen137 by homology cloning. Sequence analysis indicated that the full-length cDNAs was composed of 1 626 bp sequences including an open reading frame (ORF) of 1 521 bp region which encoded 506 amino acids. The ZmDwarf4 gene shared an amino acid sequence homology of 95%with sorghum, 89%with rice, and 71%with Arabidopsis thaliana. Real-Time PCR analysis showed that the expression level of ZmDwarf4 gene gradually in-creased from 3-leaf to 11-leaf stages with the highest expression level at 9-leaf to 11-leaf stage, the expression was gradually reduced from 12-leaf to 19-leaf stages. The expression pattern was in accordance with and the leaf angle variation of Shen137 in the period of growth and development, inferring that the ZmDwarf4 gene positive regulation the leaf angle of maize;As for the different organs, the expression level of the gene was the highest in leaf blade and lowest in pulvinus. The results of different phytohormone treatments showed that the expression of ZmDwarf4 gene of the sample which was treated by IAA was always higher than that of the control;the expression trend of BR processing samples was consistent with the control from 7-leaf to 10-leaf stages but always lower than that of the control, except that at 6-leaf stage;and the treatment with IAA+BR samples, the gene expression trend were basically the same with the control, but were higher than the control. ZmDwarf4 involved in BR biosynthesis pathway of maize to regulate the plant morphogenesis.
