Comparative Study on Surface Sterilization Methods and Viability Determination of Two Epiphytic Orchid Seeds from Gaoligong Mountains
In order to identify a convenient and efficient surface sterilization method for epiphytic orchid seeds,two species of epiphytic orchid seeds from Gaoligong Mountains were subjected to different surface sterilization meth-ods.By comparing the consistency between the TTC staining rate and asymbiotic germination rate of the seeds.The results showed that the two species of epiphytic orchid seeds had different tolerances to sterilization agents in terms of type and concentration.The seeds of Dendrobium hookerianum exhibited good tolerance to sterilization,belonging to the tolerant type of seeds,as they maintained good TTC staining rate and asymbiotic germination rate after treat-ment with 4 sterilization agents for a certain period of time.The seeds of Vanda longitepala were classified as non-tolerant seeds to sterilization.The staining rate of the seeds decreased after treatment with 4 sterilization agents for a certain period of time,but they maintained a good asymbiotic germination rate after treatment with 2%NaClO and 1%NaDCC.Based on comprehensive evaluation,treating both species of epiphytic orchid seeds with 1%NaDCC for 10-40 min showed no significant difference in germination rate,making it the optimal sterilization method for both species.
