Mitochondrial Genome Characterization and Phylogenetic Analysis of Taiwanofungus gaoligongensis
In order to understand the characteristics and phylogeny of mitochondrial genome of T.gaoligongensis,high-throughput sequencing technology was used to sequence,assemble and annotate the mitochondrial genome of T.gaoligongensis,and its composition characteristics were compared with those of other Polyporales fungi.The re-sults showed that the complete mitochondrial genome of T.gaoligongensis was obtained,and its size was 110713 bp.A total of 82 genes were encoded(53 protein coding genes,27 tRNA genes,2 rRNA genes),and there was a sig-nificant A-T preference.By collinear analysis of mitochondrial genomes of 11 closely related species,32 homologous regions were found in the mitochondrial genome of T.gaoligongensis and other mitochondrial genomes.The relative locations of homologous regions varied greatly among the Polyporales species,and frequent gene rearrangement oc-curred during the evolutionary process.Selection pressure analysis,showing Ka/Ks<1 for 15 core coding genes,and the purification selection of each species played a leading role at the amino acid level.Phylogenetic analysis showed that mitochondrial genome of T.gaoligongensis was closely related to T.camphoratus,forming an independent branch.The results showed that the mitochondrial genome of T.gaoligongensis had no change in gene sequence compared with that of T.camphoratus,but a fragment of 4209 bp was missing,which supported the analysis of T.gaoligongensis as an independent new species.
