Effect of Seed Soaking and Temperature Increase on Seed Germination and Endogenous Hormone Content of Cyclobalanopsis gilva Seeds
In order to analyse the changes of germination rhythms and endogenous hormone contents under soaking and warming treatments,and to explore the mechanism of breaking dormancy of Cyclobalanopsis gilva seeds,a two-factor analytical factorial experimental design was adopted to conduct germination tests on the seeds of C.gilva by soaking and warming treatments,and the germination situation was counted and the endogenous hormone contents of seeds were measured regularly.The results show:The highest germination rate of seed and the shortest time to reach the highest germination potential were achieved by the combination of temperature-increasing sowing and soaking seeds for 48h,and the germination time of seeds was advanced under the temperature-increasing sowing treatment. Different endogenous hormones responded differently to imbibition and warming,where warming sowing in combina-tion with imbibition reduced the content of growth hormone(IAA)in the seeds at the end of the germination period,which contributed to the germination of C.gilva seeds.The content of gibberellin(GA3)showed a fluctuating upward trend at different sowing temperatures;the content of trans-zeatin(TZR)showed a decreasing and then increasing trend under the greenhouse sowing treatment,while it showed an increasing and then decreasing trend under the warming sowing treatment;the content of jasmonic acid(JA)showed a decreasing trend under the warming treat-ment;and the content of salicylic acid(SA)showed a decreasing trend in the end of April,which was the highest for the 48 hours of soaking,and the lowest for the 24 hours of soaking,at the different sowing temperatures.The content of TZR was basically unaffected by the soaking treatments. The increase in the ratios of GA3/ABA and TZR/ABA promoted the lifting of dormancy of C.gilva acid seeds.Correlation analysis showed that TZR and GA3 were signifi-cantly positively correlated and TZR and SA were significantly negatively correlated in March,and the correlation between the hormones decreased significantly in April.Warming treatments help seed germination of C.gilva,and in conjunction with a 48-hour soak breaks seed coat boundaries,improves seed germination,and advances the time at which seed germination reaches maximum germination potential.
