Objective:To observe the effect of Jingang Pill on the expression of nuclear factor kappa-B(NF-κB)subunit(p65)protein and the content of serum Interleukin-1(IL-1)and Interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in castrated osteoporosis rats.Methods:A total of 56 SD rats were selected and divided into normal control group(NtC n=8),sham operation group(SHAM n=8)and castration group(OVX n=40).A rat model of postmenopausal osteoporosis was established by removing both ovaries of female rats in the castrated rats group.45 rats in this group were randomly divided into model group,estradiol valerate group,Jingang Pill groups of low,medium and high doses,8 rats each group.The high,medium and low dose groups of Jingang Pill and estradiol valerate group were given drug treatment for 12 weeks,once a day,and the sham operation group,normal control group and model group were given the same volume of distilled water.After 12 weeks of administration,HE staining was used to observe the morphology of bone tissue,ELISA was used to detect the contents of serum IL-1,IL-6 and TNF-α,and Western Blot was used to compare and analyze the expression of NF-кB p65 in rat femur.Results:The HE staining showed that the morphology of trabecular bone in the model group was blurred and extensively frac-tured,the bone marrow cavity was widened,and the arrangement of osteocytes was disordered.The morphology of bone tissue in the estradiol valate group and the high,medium and low dose Jingang Pill groups was significantly im-proved,the trabecular bone structure was tight,and the outer bone membrane was arranged neatly.Compared with that of the normal control group and sham operation group,the serum content of IL-1,IL-6 and TNF-α in model group were significantly increased(P<0.05),and the level of NF-κB p65 in femur was significantly increased(P<0.05).Compared with that of the model group,the content of IL-1,IL-6 and TNF-α in serum of the low,me-dium and high dose groups were significantly decreased(P<0.05),and the level of NF-κB p65 in femur was signif-icantly decreased(P<0.05).Conclusion:Jingang Pill can effectively improve the bone morphology,inhibit the pro-duction of IL-1,IL-6 and TNF-α,thereby affecting the NF-кB signaling pathway,restricting the expression ac-tivity of NF-κB p65,inhibiting the activation of osteoclasts and preventing postmenopausal osteoporosis.
