三宝护童羹对变应性鼻炎幼鼠模型细胞免疫机制的影响研究

目的 通过免疫因子及蛋白表达水平变化探讨三宝护童羹制剂(SBHTG)对卵蛋白(OVA)致SD幼鼠过敏性鼻炎(AR)肺脾气虚型模型的治疗作用及机制研究.方法 将32只2-3周龄SPF级SD大鼠分为A组:空白组,B组:模型组,C组:西药组,D组:中药组,每组各8只.适应性喂养5d后,除A组给予生理盐水模拟造模外,其余各组给予全身致敏+局部致敏法共计21d,自第15d起开始给药物干预,14d后处理.通过HE染色法评价鼻黏膜的病理形态学,以ELISA检测鼻炎相关细胞因子水平变化,取脾脏运用Western blot法检测炎症相关蛋白水平变化.结果 与A组比较,B组幼鼠鼻部症状评分、血清IL-4及IgE含量显著升高(P<0.01),三宝护童羹制剂及氯雷他定干预后显著回调;血清中INF-γ含量及脾脏中Foxp3蛋白表达显著降低(P<0.01),三宝护童羹制剂及氯雷他定干预后显著上升.C、D组幼鼠鼻部症状评分、血清Ig E及IL-4水平下降明显(P<0.01);C、D组幼鼠血清INF-γ含量显著降低(P<0.01);C、D组脾脏组织中Foxp3蛋白表达显著升高(P<0.01);C、D组鼻黏膜上皮纤毛较模型组整齐连续,上皮水肿情况、杯状细胞及EOS浸润较B组减轻.结论 推断三宝护童羹可有效调控变应性鼻炎模型幼鼠免疫功能,这可能与上调INF-γ水平及Foxp3蛋白表达水平,下调IL-4、IgE水平有关.
Effect of Sanbao Hutong Decoction on Cellular Immune Mechanism of Allergic Rhinitis In Young Rat Model
Objective:To study the therapeutic effect and mechanism of Sanbao Hutong Decoction(SBHTG)on allergic rhinitis(AR)model of qi deficiency of lung and spleen in SD young mice induced by OVA through the chan-ges of immune factors and protein expression levels.Methods:32 SPF SD rats aged 2-3 weeks were divided into blank group A,model group B,western medicine group C and Chinese medicine group D,8 rats each group.After 5 days of adaptive feeding,except for group A,the other groups were given systemic sensitization+local sensitization method for 21 days,drug intervention began on the 15th day,and the treatment was given 14 days later.The pathologi-cal morphology of nasal mucosa was evaluated by HE staining,the changes of rhinitis related cytokines were detected by ELISA,and the changes of inflammatory related proteins were detected by Western blot.Results:Compared with those of group A,the nasal symptom score,serum IL-4 and IgE contents were significantly increased in group B(P<0.01),and significantly decreased after intervention with Sanbao Hutong Decoction and loratadine.INF-γ content in serum and Foxp3 protein expression in spleen were significantly decreased(P<0.01),and were significantly in-creased after intervention with Sanbao Hutong Decoction and loratadine.Nasal symptom scores,serum IgE and IL-4 levels of the rats in groups C and D were significantly decreased(P<0.01).Serum INF-γ content in groups C and D was significantly decreased(P<0.01).The expression of Foxp3 protein in spleen tissues of groups C and D was significantly increased(P<0.01).The nasal mucosal cilia in groups C and D were more orderly and continuous than those in the model group,and the epithelial edema,goblet cells and EOS infiltration were less severe than those in group B.Conclusion:It is concluded that Sanbao Hutong Decoction can effectively regulate the immune function of young mice with allergic rhinitis model,which may be related to up-regulating INF-γ level and Foxp3 protein ex-pression level,and down-regulating IL-4 and IgE level.
