Exploration of the Mechanism of Paeoniae in the Treatment of Non-alcoholic Fatty Liver Disease Based on Network Pharmacology
Objective:To explore the mechanism of Paeoniae in the treatment of non-alcoholic fatty liver disease by using the method of network pharmacology. Methods:The effective components and main component targets of Pae-oniae were screened by TCMSP and Swiss Target Prediction database. The target gene of the disease was found by u-sing Genecards database, and the intersection map of Paeoniae component target and disease target was obtained by Venny2.1 online mapping platform. And then a network of Paeoniae-active-component-target was constructed by Cytoscape3.7.2 software and PPI protein network and DAVID database were constructed by String database to make gene GO analysis and KEGG pathway analysis. Results:A total of 13 effective components,264 drug target genes,950 disease target genes and 83 intersection target genes were obtained through screening. The gene GO analysis showed that Paeoniae was mainly involved in the positive regulation of RNA polymerase Ⅱ promoter transcription, the negative regulation of apoptosis, protein binding and other biological processes. The KEGG pathway analysis showed 20 closely related pathways, including cancer pathway,HIF-1 signaling pathway,MAPK signaling pathway,PI3K-Akt signaling pathway, lipid and atherosclerosis. Conclusion:Paeoniae exerts its role in the treatment of non-alcohol-ic fatty liver disease through multi-target and multi-pathway.
