Objective:To study the effect of Sini Decoction on the proliferation and apoptosis of A549 lung cancer stem cells.Methods:A549 lung cancer stem cells were induced by serum-free suspension culture,and the positive rate of CD133 was detected to verify the cell stemness.The A549 lung cancer stem cells were intervened with different doses of herb-containing serum of Sini Decoction(10％,20％).The effect of Sini Decoction on the proliferation and apoptosis of A549 lung cancer stem cells was detected by CCK-8 and flow cytometry.And qPCR was used to detect the effect of Sini Decoction on the proliferation and apoptosis of A549 lung cancer stem cells.Results:The positive rate of CD133 was(82.3667±0.47258)％,which was significantly higher than that of lung cancer cell group(P＜0.05).After CCK-8 detection,the light absorption value of lung cancer stem cells in the Sini Decoction intervention group was significantly decreased compared with that of the control group at 48 and 72h(P＜0.05),and the apoptosis rate of lung cancer stem cells in the Sini Decoction intervention group was significantly increased compared with that of the control group(P＜0.05).The herb-containing serum of Sini Decoction significantly decreased the mRNA levels of PCNA,Ki67 and Bcl-2 of A549 lung cancer stem cells(P＜0.05),and significantly increased the mRNA levels of Bax of lung cancer stem cells(P＜0.05).Conclusion:Sini Decoction can inhibit the proliferation and promote the apoptosis of A549 lung cancer stem cells.
