Study on the Anti-depression Mechanism of Tibetan medicine Shi-Yi-Wei-Wei-Ming capsule Based on BDNF/CREB Signaling Pathway
Objective:To study on the anti-depression effect and mechanism of Tibetan medicine Shi-Yi-Wei-Wei-Ming capsule on depressed mice based on BDNF/CREB signaling pathway.Methods:48 mice were randomly divided into normal group(n=12)and model group(n=36).CUMS combined with CRS method was used to make the model group for 3 weeks.After modeling,the mice were randomly divided into model group,Shi-Yi-Wei-Wei-Ming capsule group(Tibetan medicine group)and fluoxetine group,12 mice each group,and the intervention lasted for 4 weeks.Behavioral experiments were used to evaluate the depression mouse model.The expression of BDNF,GDNF,NGF,p-CREB/CREB and PSD-95 in hippocampus of mice were detected by WB method.The MOD values of BDNF,GDNF and p-CREB were determined by IHC.Results:After 3 weeks of modeling,the mice in the model group showed depression-like behavior,and the mice body mass and sucrose preference rate were lower than those in the normal group(P<0.01).The immobility time was longer than that of the normal group(P<0.01).After 4 weeks of adminis-tration,the body mass and sucrose preference rate of mice in the model group were lower than those in the normal group(P<0.01).The immobility time was longer than that of the normal group(P<0.01).The body mass and sucrose pref-erence rate of mice in the Tibetan medicine group and fluoxetine group were higher than those in the model group(P<0.05 or P<0.01).The immobility time was shorter than that of the model group(P<0.01).The expressions of BDNF,GDNF,PSD-95 protein and the MOD values of BDNF,GDNF and P-CREB in the hippocampus of mice in the model group were lower than those in the normal group(P<0.05 or P<0.01).The protein expressions of BDNF,GDNF,P-CREB/CREB and PSD-95 in the hippocampus of mice in the Tibetan medicine group and the fluoxetine group were higher than those in model group(P<0.05 or P<0.01).The MOD values of BDNF and GDNF in the Tibetan medicine group were higher than those in the model group(P<0.05 or P<0.01).The hippocampal BDNF,GDNF and P-CREB MOD values of the fluoxetine group were higher than those of the model group(P<0.01).Conclusion:Shi-Yi-Wei-Wei-Ming capsule can improve the depressive behavior of depressed mice,and its mechanism may be related to the regulation of BDNF/CREB signaling pathway.
