摘要
目的 运用网络药理学探讨秦国政教授"秦氏四妙散"治疗慢性前列腺炎的潜在作用机制.方法 通过TCMSP收集药物活性成分及靶标,利用Cytoscape3.8.0构建"药物-活性成分-靶点"网络图;在OMIM、Gene-cards、Drugbank等多个数据库中以"chronic prostatitis"为关键词检索疾病相关靶点,利用String数据库获取PPI网络图并进行拓扑分析.运行G0和KEGG富集分析,使用R4.0.3软件,绘制气泡图、柱状图.结果 秦氏四妙散共含144个活性成分,潜在作用靶点有138个,可筛选出MAPK1、TP53、STAT3等11个核心靶标.G0富集结果共2645条;KEGG结果涉及179条信号通路.网络药理学分析结果表明,槲皮素、山柰酚、甘草查尔酮A、柚皮素等是秦氏四妙散的主要活性成分,STAT3、TP53、MAPK1等是秦氏四妙散治疗慢性前列腺炎的主要靶点,共同靶点富集于PI3K-Akt信号通路、AGE-RAGE信号通路、IL-17信号通路、TNF信号通路等多个信号通路,调节细胞、凋亡、基因转录等生物学过程.结论 本研究阐述了秦氏四妙散治疗慢性前列腺炎"多成分、多通路、多靶点"的可能机制,为进一步了解秦氏四妙散的作用机制,探索新的临床应用和实验探究提供了一定的理论依据和参考.
Abstract
Objective:To explore the possible mechanism of action of"Qin's Four Wonders Powder"by Professor Qin Guozheng in the treatment of chronic prostatitis by using network pharmacology.Methods:Herbal active ingredi-ents and targets were collected by TCMSP,and the"herb-active ingredient-target"network diagram was construc-ted using Cytoscape3.8.0.Disease-related targets were retrieved with the keyword"chronic prostatitis"in multiple databases such as OMIM,Genecards,and Drugbank.The PPI network graph was obtained using the String database and topological analysis was conducted.G0 and KEGG enrichment analyses was performed,and R4.0.3 software was used to draw bubble charts and bar charts.Results:Qin's Four Wonders Powder contained a total of 144 active ingre-dients,with 138 potential targets.Eleven core targets such as MAPK1,TP53,and STAT3 could be screened out.A total of 2,645 G0 enrichment results were obtained.The KEGG results involved 179 signaling pathways.The results of network pharmacological analysis showed that quercetin,kaempferol,glycyrrhizin A,naringin,etc.were the main ac-tive ingredients of Qin's Four Wondwrs Powder,while STAT3,TP53,MAPK1,etc.were the main targets of Qin's Four Wonders Powder in the treatment of chronic prostatitis.The common targets were enriched in multiple signaling path-ways such as the PI3K-Akt signaling pathway,the AGE-RAGE signaling pathway,the IL-17 signaling pathway,and the TNF signaling pathway and regulating biological processes such as cells,apoptosis,and gene transcription.Conclusion:This study expounded the action mechanism of"multiple components,multiple pathways,and multiple targets"of Qin's Four Wonders Powder in the treatment of chronic prostatitis,providing a certain theoretical basis and reference for further understanding the mechanism of action of Qin's Four Wonders Powder and exploring new clinical applications and experimental research.
