基于网络药理学探讨海藻治疗乳腺增生的作用机制

目的 运用网络药理学探究海藻治疗乳腺增生的作用机制。方法 通过TCMSP数据库获取、筛选海藻的主要活性成分及相对应的作用靶点;通过数据库检索获取乳腺增生疾病相关靶点,构建蛋白互作(PPI)网络并分析,获取核心作用靶点;进行功能富集分析,并构建海藻治疗乳腺增生的"活性成分-靶点-通路"网络图。结果 海藻中的槲皮素等活性成分,以及核心靶点热休克蛋白 90α家族A类成员 1(HSP90AA1)、丝裂原激活的蛋白激酶 1(MAPK1)、肿瘤蛋白p53(TP53)、表皮生长因子受体(EGFR)、肿瘤坏死因子(TNF)与海藻治疗乳腺增生相关,主要参与调控细胞对化学应激的反应、膜微域和膜阈等生物过程,通过调节癌症、脂质和动脉粥样硬化等信号通路产生治疗乳腺增生的作用。结论 应用网络药理学初步揭示了海藻治疗乳腺增生的作用机制,为其临床运用提供了理论支撑。
Exploring the Mechanism of Sargassum on Hyperplasia of Mammary Glands through Network Pharmacology
Objective To study the mechanism of sargassum in the treatment of hyperplasia of mammary glands(HMG)through network pharmacology.Methods The active constituents and targets of sargassum were searched and screened from TCMSP.The targets of HMG were obtained through database search.The PPI network was constructed for interaction analysis,and obtain core targets.Functional enrichment analysis was performed on the core targets.The network diagram of"component-target-pathway"of sargassum in treating HMG was constructed.Results The quercetin of sargassum and the core targets,such as HSP90AA1,MAPK1,TP53,EGFR and TNF were related to the treatment of HMG,which mainly involved in cellular response to chemical stress,membrane microdomain,membrane raft,mainly enriched in pathways in cancer,lipid and atherosclerosis.Conclusion This study has initially reveal the mechanism of treating HMG with sargassum through network pharmacology,which can provide reference for clinical application.
