摘要

目的 建立超高效液相色谱-二极管阵列检测器(UPLC-DAD)法测定人参再造丸中芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚 5 种成分的含量.方法 采用Waters CORTECS UPLC C18 色谱柱(2.1 mm×100 mm,1.6 μm);以甲醇-0.1%磷酸溶液为流动相,梯度洗脱,流速为 0.3 mL/min;检测波长为 437 nm;柱温 40℃.结果 芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚 5 种成分分离度良好,在相应范围内线性关系良好,相关系数(r)均不低于0.999 7,平均回收率分别为 92.5%、83.5%、94.3%、98.9%、106.8%,RSD(n=6)分别为 2.0%、1.2%、2.6%、2.4%、2.4%.结论 UPLC-DAD法准确可靠、重复性好,可用于人参再造丸的质量控制.

Abstract

Objective To establish ultra high performance liquid chromatography-diode array detector(UPLC-DAD)method to determine the content of aloe emodin,rhein,emodin,chrysophanol and physcione in renshen zaizao pills.Methods Adopts Waters CORTECS UPLC C18 column(2.1 mm×100 mm,1.6 μm),with methanol(A),0.1%phosphate water(B)as the mobile phase,gradient elution,the flow rate was 0.3mL/min;the detection wavelength was 437 nm;the column temperature was 40℃.Results Aloe emodin,rhein,emodin,chrysophanol and physcione had good separation,with good linear relationship in the corresponding range,and the correlation coefficient(r)was not lower than 0.999 7,the average recovery rate was 92.5%,83.5%,94.3%,98.9%and 106.8%,and RSD(n=6)was 2.0%,1.2%,2.6%,2.4%and 2.4%,respectively.Conclusion The method used is accurate and reliable,and can be used for the quality control of renshen zaizao pills.
